维生素D对发生氧化损伤的猪睾丸间质细胞保护作用的研究 被引量:2

摘要 为了探究维生素D对发生氧化损伤的猪睾丸间质细胞是否具有保护作用,试验利用H_(2)O_(2)使间质细胞发生氧化损伤,再利用1α,25-(OH)_(2)VD_(3)刺激间质细胞,然后检测间质细胞的活力、总抗氧化能力、过氧化氢酶活力与培养液内睾酮浓度。结果表明:与不添加H_(2)O_(2)时相比,培养液内添加500μmol/L的H_(2)O_(2)可使间质细胞活力、总抗氧化能力、过氧化氢酶活力与睾酮分泌能力极显著降低(P<0.01),500μmol/L H_(2)O_(2)可使间质细胞发生氧化损伤。当在培养液内同时添加500μmol/L的H_(2)O_(2)与100 nmol/L的1α,25-(OH)_(2)VD_(3)时,与仅添加500μmol/L的H_(2)O_(2)相比,间质细胞的活力、总抗氧化能力、过氧化氢酶活力与睾酮分泌能力均极显著提高(P<0.01)。说明维生素D对发生氧化损伤的猪睾丸间质细胞具有保护作用。 In order to explore the protective effect of vitamin D on porcine leydig cells with oxidative damage, H_(2)O_(2)was used to induce oxidative damage to the leydig cells, and 1α,25-(OH)_(2)VD_(3)was used to stimulate the leydig cells. The activity of leydig cells, total antioxidant capacity, catalase activity and testosterone concentration in culture medium were detected. The results showed that compared with no H_(2)O_(2), 500 μmol/L of H_(2)O_(2)could significantly reduce the interstitial cell activity, total antioxidant capacity, catalase activity and testosterone secretion capacity of interstitial cells(P<0.01), and 500 μmol/L of H_(2)O_(2)could cause oxidative damage of interstitial cells. When 500 μmol/L of H_(2)O_(2)and 100 nmol/L of 1α,25-(OH)_(2)VD_(3)were added simultaneously in culture medium, the interstitial cell activity, total antioxidant capacity, catalase activity and testosterone secretion capacity of interstitial cells were significantly increased compared with the addition of 500 μmol/L of H_(2)O_(2)(P<0.01). These results indicated that vitamin D had protective effect on porcine leydig cells with oxidative damage.
