摘要
目的:探讨通督调神针刺改善脑缺血再灌注损伤大鼠神经功能机制。方法:选取健康雄性SD大鼠40只随机分为假手术组、模型组、药物组、针刺组各10只,采用改良Koizumi线栓法建立大鼠脑缺血再灌注损伤模型,缺血2 h再灌注24 h后,药物组大鼠腹腔注射丁苯酞氯化钠注射液,针刺组大鼠实施通督调神针刺治疗7 d,治疗前后均计算大鼠神经功能缺损评分(neurological severity scores,NSS),采用免疫组化和Western blot方法测定血管内皮生长因子(vascular endothelial growth factor,VEGF)、神经生长因子(nerve growth factor,NGF)、髓鞘碱性蛋白(myelin basic protein,MBP)表达。结果:药物组、针刺组治疗后NSS评分均低于治疗前(P<0.05);药物组治疗后NSS评分低于模型组(P<0.05),针刺组治疗后NSS评分低于模型组和药物组(P<0.05);药物组脑缺血组织中的VEGF、NGF、MBP表达高于模型组(P<0.05),针刺组脑缺血组织中的VEGF、NGF、MBP表达高于模型组和药物组(P<0.05)。结论:通督调神针刺明显改善脑缺血再灌注损伤大鼠的神经功能,其机制可能与针刺上调VEGF、NGF、MBP表达进而营养神经、促进神经髓鞘组织再生有关。
Objective:To explore the mechanism of Tongdu Tiaoshen acupuncture on improving nerve function in rats with cerebral ischemia reperfusion injury.Methods:Forty healthy male SD rats were enrolled and randomly divided into sham operation group,model group,drug group and acupuncture group,10 cases in each group.CI-RI rats models were established by modified Koizumi suture method.After 2h of ischemia and 24h of reperfusion,drug group was intraperitoneally injected with butylphthalide sodium chloride injection,while acupuncture group was given Tongdu Tiaoshen acupuncture for 7 days.Neurological Severity Scores(NSS)were calculated before and after treatment.The expression of vascular endothelial growth factor(VEGF),nerve growth factor(NGF)and myelin basic protein(MBP)was measured by immunohistochemistry and Western blot.Results:After treatment,the NSS of drug group and acupuncture group were decreased(P<0.05).The NSS of drug group was lower than that of model group(P<0.05),and the NSS of acupuncture group was lower than that of model group and drug group(P<0.05).The expressions of VEGF,NGF and MBP of cerebral ischemic tissues in drug group were higher than those in model group(P<0.05).The expressions of VEGF,NGF and MBP in acupuncture group were higher than those in model group and drug group(P<0.05).Conclusion:Tongdu Tiaoshen acupuncture can significantly improve nerve function of CI-RI rats.The mechanism may be related to that acupuncture up-regulates the expression of VEGF,NGF and MBP,nourishing nerves and promotes regeneration of nerve myelin tissue.
