摘要
采用响应面法优化海洋放线菌DUT11产衣霉素的发酵工艺。在单因素试验基础上,选择可溶性淀粉的含量、黄豆粉的含量和发酵时间为试验因素,进行三因素三水平Box-Behnken中心组合设计。实验得到最佳发酵工艺:4%可溶性淀粉,2.5%黄豆粉,0.25%酵母粉,0.25%的K_(2)HPO_(4)和0.3%CaCO_(3),发酵温度28℃,初始pH值为7,发酵6d。最终得到衣霉素的实际产量为53.15mg·L^(-1),预测值为52.98mg·L^(-1),理论值与实验值很接近。结果证明,该响应面优化工艺合理、可行。
Response surface analysis methodology(RSM)was applied to optimize the fermentation conditions of marine actinomycete DUT11 for the production of tunicamycin.Base on the single-factor experiments,three-factors-three-levels Box-behnken central component experiments design were applied.The concentrations of soluble starch,soybean flour and fermentation time were selected as the experimental factors.The optimum fermentation conditions were as follows:4%soluble starch,2.5%soybean powder,0.25%yeast powder,0.25%K_(2)HPO_(4)and 0.3%CaCO_(3),fermentation temperature 28℃,initial pH 7,culture time 6 days.Finally,the actual yield of tunicamycin was 53.15mg·L^(-1),the predicted value was 52.98mg·L^(-1),and the theoretical value was close to the experimental value.These results demonstrated that this method is feasible.
