摘要
目的研究毒胡萝卜素(Thapsigargin)对PC12细胞的影响。方法将体外常规培养的PC12细胞分为正常组和Thapsigargin组,利用Thapsigargin诱导PC12细胞凋亡,建立缺血后神经元损伤模型。采用MTT法检测存活率,采用Annexin V-FITC/PI法检测凋亡率情况。结果Thapsigargin组吸光度A值为(0.1497±0.0357),低于正常组的(0.2573±0.0093),差异有统计学意义(P<0.05);Thapsigargin组早期凋亡率及总凋亡率高于正常组,差异有统计学意义(P<0.05);Thapsigargin组JNK3、Caspase-3蛋白表达高于正常组,差异有统计学意义(P<0.05)。结论JNK3、Caspase-3可能参与了神经细胞的损伤过程。
Objective To investigate the effects of thapsigargin on PC12 cells.Methods PC12 cells cultured in vitro were divided into normal group and Thapsigargin group;Thapsigargin was used for inducing the apoptosis of the PC12 cells,and the model of neuronal injury after ischemia was established.MTT assay was used for measuring the cell viability and Annexin V-FITC/PI was used for measuring the apoptosis rate.Results The absorbance A value of Thapsigargin group was(0.1497±0.0357),which was lower than that of normal group(0.2573±0.0093),and the difference was statistically significant(P<0.05).The early apoptosis rate and total apoptosis rate in the Thapsigargin group were higher than those in the normal group,and the difference was statistically significant(P<0.05).The expression of JNK3 and Caspase-3 protein in Thapsigargin group were higher than that in normal group,and the difference was statistically significant(P<0.05).Conclusion JNK3 and Caspase-3 may be involved in neuronal injury.
