系统性红斑狼疮患者尿液IFN-α及TRP-KYN通路与临床特征的相关性研究

摘要 系统性红斑狼疮(systemic lupus erythemato-sus,SLE)是自身免疫介导的以免疫性炎症为突出表现的弥漫性结缔组织病,血清中多种自身抗体的产生和多系统受累是SLE的两个主要特征[1]。临床上,尿液标志物的评估常常被忽略。犬尿氨酸(kynure-nine,KYN)是吲哚胺2,3-双加氧酶(indoleamine 2,3-dioxygenas,IDO)作用下的色氨酸(tryptophan,TRP)代谢产物。干扰素α(interferonα,IFN-α)等炎症因子可影响IDO活性,造成TRP-KYN通路失衡。研究表明,SLE等自身性免疫疾病中伴有抑郁、焦虑和认知缺陷患者的KYN/TRP比值存在异常[2]。SLE患者血清色氨酸浓度较正常人降低,KYN水平和IDO活性较正常人显著增高[3]。TRP-KYN通路是免疫系统和神经系统之间相互影响的关键界面。IFN-α在SLE的发病中起着至关重要的作用[4],SLE患者出现精神异常的分子机制很可能与IFN-α等炎症因子诱导的TRP-KYN通路异常有关。因此,为了寻求尿液中评价SLE患者的生物标志物。本研究应用酶联免疫吸附测定(ELISA)分析SLE患者尿液IFN-α及TRP-KYN通路与临床特征的相关性。
