26份玉米材料灰斑病抗性鉴定及分子标记检测 被引量:1

Identification and Molecular Detection of Gray Leaf Spot Resistance in 26 Elite Maize Inbred Lines

摘要 为了解不同来源玉米自交系灰斑病抗性位点分布,探索利用分子标记筛选玉米灰斑病抗性种质,本研究以26份玉米骨干自交系为材料,按照国家标准(NY/T 1248.11-2016)进行人工接种鉴定,选用6个与抗灰斑病主效QTL(或基因)紧密连锁的分子标记进行分子检测。结果表明,经接种鉴定,抗性级别表现为中抗及以上的有15份,其中高抗有4份;表现感病的有11份。结合接种鉴定和分子标记检测表明,供试材料抗病位点主要集中在KHB5、qRgls2.04、qRgls1.06、qRgls2.07和qRgls3.02等5个位点。标记umc 2614和TID 5306可以检测与齐319(高抗)一致的抗病位点(KHB5和qRgls2.04);umc 1972、umc 1042和bnlg 1523可以检测与T 32(高抗)一致的抗病位点(qRgls1.06、qRgls2.07和qRgls3.02),接种鉴定与分子检测的结果总体表现一致。研究表明,利用分子标记检测,能有效辅助筛选玉米抗灰斑病种质,对玉米的分子育种具有重要意义。 To understand the distribution resistance loci to gray leaf spot in different maize inbred lines and develop the molecular markers to screen maize germplasms with gray leaf spot resistance,26 maize backbone inbred lines in this study were used for artificial inoculation following the national standard(NY/T 1248.11-2016),and six known molecular markers closely linked to the major QTL(or gene)for gray spot resistance were chosen for molecular detection of these materials.As a result,after inoculated identification,15 inbred lines showed medium or above resistance,including 4 germplasms with high resistance and 11 germplasms with susceptibilty.Combined with inoculated identification and molecular marker detection showed that the disease-resistant loci of the tested materials were mainly concentrated in five sites,namely KHB5,qRgls2.04,qRgls1.06,qRgls2.07 and qRgls3.02.Markers umc 2614 and TID 5306 could detect disease resistance loci(KHB5 and qRgls2.04)consistent with highly resistant Qi 319(high resistance).And umc 1972,umc 1042 and bnlg 1523 could detect disease resistance loci(qRgls1.06,QRgls2.07 and qRgls3.02)consistent with T 32(high resistance),the overall performance of the vaccination identification and molecular testing results were consistent.Therefore,useing of molecular marker detection could effectively assist the screening of maize germplasm resistant to gray spot disease,which was of great significance to molecular breeding in maize.
