摘要
目的研究子宫内膜异位症(EMs)患者子宫内膜细胞的生化代谢特性,探讨其在疾病发生发展中的作用。方法收集EMs患者异位及在位子宫内膜组织、对照组子宫肌瘤患者子宫内膜组织。利用单细胞激光光镊拉曼光谱系统,测定并比较不同组别子宫内膜细胞的拉曼光谱。结果3组细胞拉曼光谱波峰位置及形态大致相似,部分波峰强度有差异。与患者在位子宫内膜和对照组子宫内膜相比,异位子宫内膜细胞内胆固醇(702cm^(-1))、类胡萝卜素(1161、1525cm^(-1))、核酸(1094cm^(-1))等成分减少,而蛋白(758、831、853、1209、1245、1589cm^(-1))、脂类(1132cm^(-1))、糖原(492cm^(-1))相关的成分增加。通过PCA聚类分析可以看出,内异症患者在位子宫内膜细胞与对照组子宫内膜细胞光谱相似性较高,而异位子宫内膜细胞中出现离群细胞。结论EMs患者异位子宫内膜细胞分子代谢水平发生了较大的改变,可能与疾病的发生发展过程有关。
Objective:To study the biochemical and metabolic properties of endometrial cells in endometriosis patients and investigate its role in the development of the disease.Methods:The ectopic or eutopic endometrial tissue was collected from endometriosis patients and control group patients with uterine fibroids.Measured by single-cell Raman trapping microspectroscopy,the Raman spectra of ectopic or eutopic endometrial cells from different groups were analyzed and compared.Results:The shapes and peak positions of Raman spectra from the three groups were roughly similar,but some of the peak intensities were different.Compared with the patient's eutopic endometrium and the control endometrium,the ectopic endometrium showed decreased intracellular cholesterol(702cm^(-1)),carotenoids(1161,1525cm^(-1))and nucleic acids(1094cm^(-1)),while increased protein(758,831,853,1209,1245,1589cm^(-1)),lipids(1132cm^(-1))and glycogen(492cm^(-1)).Through PCA cluster analysis,it could be seen that the spectra of eutopic endometrial cells of endometriosis patients were similar to those of the control group,while some ectopic endometrial cells were far away from them.Conclusions:The molecular metabolism of ectopic endometrium cells in patients with endometriosis was significantly changed,which may be related to the occurrence and development of the disease.
