丹参、泽泻对NAFLD大鼠脂代谢及PPARα基因表达的影响 被引量:9

摘要 目的:探究丹参、泽泻对非酒精性脂肪肝(NAFLD)的可能作用机制。方法:将60只Wista大鼠随机分为6组:正常组、模型组、东宝甘泰组和丹参水煎剂组、泽泻水煎剂组、丹参-泽泻(丹泽)水煎剂组。通过喂饲高脂饲料建立NAFLD模型,同时给予相应药物治疗。8周后,检测各组大鼠血清胆固醇(TC)、甘油三酯(TG)、游离脂肪酸(FFA)、谷草转氨酶(AST)和谷丙转氨酶(ALT)水平及过氧化物酶体增殖物激活受体α(PPARα)mRNA表达,HE法观察大鼠肝组织病理形态学改变。结果:各用药组均能明显改善血脂和肝功能(P<0.01),升高肝组织PPARαmRNA的表达(P<0.01),尤以丹泽组降脂作用更为明显。结论:丹参、泽泻和丹泽水煎剂能显著改善NAFLD大鼠脂代谢紊乱,改善肝功能,其机制可能与增强肝组织中PPARαmRNA表达有关。 Objective:To explore the possible mechanism of Danshen(Salvia miltiorrhiza)and Zexie(Alisma orientalis)on nonalcoholic fatty liver disease(NAFLD).Methods:60 Wista rats were randomly divided into 6 groups:normal group,model group,Dongbao Gantai group,Danshen Decoction group,Zexie Decoction group and Dan-ze(Danshen-Zexie)Decoction group.NAFLD model was established by feeding high-fat diet,and corresponding medicinal treatment was given at the same time.After 8 weeks,the levels of serum cholesterol(TC),triglyceride(TG),free fatty acids(FFA),aspartate aminotransferase(AST),alanine aminotransferase(ALT)and peroxisome proliferator activated receptorα(PPARα)were measured;The pathomorphological changes of rat liver were observed by HE method.Results:All treatment groups could significantly improve blood lipid and liver function(P<0.01),and increase PPARαmRNA expression in liver tissue(P<0.01),especially in Dan-ze group.Conclusion:Danshen,Zexie and Dan-ze decoction can significantly improve the disorder of lipid metabolism and liver function in NAFLD rats.The mechanism may be related to the enhancement of PPARαmRNA expression in liver tissue.
