MAC30表达水平对非小细胞肺癌患者化疗效果及预后的影响

Influence of MAC30 Expression Level on Chemotherapy Effect and Prognosis of Patients with Non-small Cell Lung Cancer

目的探究脑膜瘤相关蛋白(MAC30)表达水平对非小细胞肺癌患者化疗效果及预后的影响。方法选取64例NSCLC患者作为研究对象,使用TC方案进行化疗。采用免疫组织化学法检测肿瘤组织及癌旁组织中MAC30的表达情况,采用RT-PCR技术检测MAC30 mRNA表达水平,分析MAC30水平与化疗效果的关系,采用Kaplan-Meier生存曲线分析MAC30水平与患者预后的关系。结果MAC30蛋白在NSCLC组织中高表达率为81.25%,明显高于癌旁组织中高表达率23.44%(P<0.001);NSCLC组织中MAC30 mRNA水平明显高于癌旁组织(P<0.001)。MAC30表达水平与NSCLC患者年龄、性别及肿瘤病理类型无关(P>0.05),与肿瘤TNM分期、有无淋巴结转移和分化程度相关(P<0.001)。不同效果组的患者组织MAC30 mRNA水平差异具有统计学意义(P<0.001),CR组及PR组患者MAC30 mRNA水平明显低于PD组患者(P<0.05)。MAC30高表达患者生存率明显低于低表达患者(P<0.05)。结论MAC30在NSCLC组织中高表达且与化疗效果和预后相关,可能作为NSCLC化疗效果及患者预后的相关评估指标。

Objective To explore the nce of the expression level of meningioma-related protein(MAC30)on the chemotherapy effect and prognosis of patients with non-small cell lung cancer.Methods 64 NSCLC patients were selected as the research subjects,and the TC regimen was used for chemotherapy.Immunohistochemistry was used to detect the expression of MAC30 in tumor tissues and adjacent tissues,RT-PCR was used to detect the expression of MAC30 mRNA,the relationship between MAC30 levels and chemotherapy effects was analyzed,and the Kaplan-Meier survival curve was used to analyze he relationship between MAC30 levels and patient prognosis.Results The high expression rate of MAC30 protein in NSCLC tissue was 81.25%,which was significantly higher than that in adjacent tissues of 23.44%(P<0.001);MAC30 mRNA level in NSCLC tissue was significantly higher than that in adjacent tissues(P<0.001).The expression level of MAC30 had nothing to do with the age,gender and tumor pathological type of NSCLC patients(P>0.05),but was related to the tumor TNM stage,the presence or absence of lymph node metastasis and the degree of differentiation(P<0.001).The differences in the levels of MAC30 mRNA in the tissues of patients in different effect groups were statistically significant(P<0.001),and the levels of MAC30 mRNA in the CR and PR groups were significantly lower than those in the PD group(P<0.05).The survival rate of patients with high MAC30 expression was significantly lower than that of patients with low MAC30 expression(P<0.05).Conclusion MAC30 is highly expressed in NSCLC tissues and is related to the effect of chemotherapy and prognosis.It may be used as a relevant evaluation index for the effect of chemotherapy and patient prognosis in NSCLC.
