新疆紫草的红外指纹图谱及高效薄层色谱研究

摘要 目的以新疆紫草作为研究对象,构建其红外指纹图谱及高效薄层色谱(HPTLC),分析其差异。方法利用红外光谱仪测定新疆紫草的红外光谱。使用OriginPro 8.5.1软件绘制新疆紫草的红外光谱图。SPSS软件进行聚类分析及相似度分析。用高效薄层色谱法对新疆紫草中4种化学成分进行定性定量分析。结果红外指纹图谱表明,各紫草相似度大于0.97。聚类分析中当欧式距离大于25时,紫草才可以分为一类。高效薄层色谱表明,左旋紫草素、β-乙酰氧基异戊酰阿卡宁、乙酰紫草素及β,β-二甲基丙烯酰紫草素的RF值分别为(0.37±0.05),(0.46±0.05),(0.57±0.05)及(0.74±0.05)。平均加标回收率为99.68%、100.21%、100.20%及98.10%。结论通过红外光谱的鉴别发现,新疆紫草中所含成分间的官能团差异较小。用高效薄层色谱法可有效分离4种成分,快速检测新疆紫草中各成分含量的差异,为其进一步研究奠定了基础。 OBJECTIVE To construct the infrared fingerprint and high performance thin layer chromatography(HPTLC)of Arnebia euchroma as a research object and analyze the differences.METHODS The infrared spectra of Arnebia euchroma were determined using an infrared spectrometer.The infrared spectra of Arnebia euchroma were plotted using OriginPro 8.5.1 software.Cluster analysis and similarity analysis were performed by SPSS software.The qualitative and quantitative analysis of the four chemical components in Arnebia euchroma was carried out by high performance thin layer chromatography.RESULTS The infrared fingerprints showed that the similarity of each comfrey was>0.97.In the cluster analysis,Arnebia euchroma could be classified into one category only when the Euclidean distance was greater than 25.The mean spiked recoveries were 99.68%,100.21%,100.20%and 98.10%for shikonin,β-acetoxyisovalerylarkanin,acetylshikonin andβ,β-dimethylacrylonylshikonin,respectively.CONCLUSION The functional groups of the components contained in Arnebia euchroma were found to be less different from each other by IR spectroscopy.HPTLC method can effectively separate the four components and rapidly detect the differences in the content of each component in Arnebia euchroma,laying the foundation for its further study.
