麦门冬汤对硬皮病模型小鼠的疗效及肺与皮肤组织相关免疫因子的影响 被引量:3

摘要 目的系统性硬化症(systemic sclerosis,SSc)的发病与免疫功能密切相关。通过观察麦门冬汤对博来霉素所致BALB/c硬皮病模型小鼠的疗效和肺、皮肤相关免疫因子的影响,探讨麦门冬汤在治疗SSc过程中对“卫气(免疫功能)”与“肺主皮毛”的影响及作用机制。方法建立硬皮病小鼠模型后,随机将60只小鼠均分为模型组、麦门冬高、中、低剂量组,并设正常对照组。治疗3周后采集标本,切取小鼠背部注射区皮肤及肺部组织,分别用以测定单位皮肤及肺组织中皮肤厚度、羟脯氨酸、胶原含量、纤维化指数、CTGF、PDGF、IL-10、TGF-β和TNF-α等的含量。结果麦门冬汤能极显著降低模型组小鼠皮肤厚度、羟脯氨酸、胶原含量、皮肤纤维化指数和肺纤维化指数(P<0.01);麦门冬汤能极显著降低模型组小鼠皮肤和肺中CTGF、PDGF、IL-10、TGF-β、TNF-α的含量(P<0.01)。结论麦门冬汤对小鼠SSc有良好的治疗效果,且存在量效关系;麦门冬汤治疗SSc的机制可能与降低羟脯氨酸和胶原的含量、皮肤和肺的纤维化指数、CTGF、PDGF、IL-10、TGF-β和TNF-α的含量有关。 Objective Incidence of systemic sclerosis is closely related to immune function.In this study,by observing the effect of Maimendong decoction and and related immune factors of lung and skin tissue in BALB/c scleroderma model mice,mechanism of the Wei,Qi(immune function)and"Lung master fur"of Maimendong Decoction treatment of SSc were investigated.Methods After scleroderma mice model was established,60 mice were randomly divided into model group,the Maimendong high dose,middle dose and low dose group and normal control group.After three weeks of treatment to collect specimens.central injection site skin of mice back and lung tissue were cut,it is used to determine the skin thickness,hydroxyproline,collagen content,fibrosis index,CTGF,PDGF,IL-10,TGF-βand TNF-αcontent in skin and lung tissue respectively.Results Maimendong decoction can reduce the skin thickness,pulmonary fibrosis index and content of hydroxyproline in the model group mice(P<0.01).(2)Maimendong decoction can reduce the content of CTGF and PDGF,IL-10 and TGF-beta and TNF-alpha in the skin and lung tissue of mice of modelgroup mice(P<0.01).Conclusion(1)Maimendong decoction on mice SSc good therapeutic effect and dose-effect relationship.(2)Mechanism of Maimendong decoction on SSc may reduce the content of hydroxyproline and collagen in skin,skin and lung fibrosis index,the content of of CTGF,PDGF,IL-10,TGF-βand TNF-α.
