基于液相质谱技术的真武汤对抗阿霉素心肌细胞的毒性机制探讨

摘要 目的基于液相质谱法的非靶向代谢组学技术研究阿霉素心肌毒性大鼠特征性代谢物变化,了解真武汤的疗效机制。方法18只雄性SD大鼠,分为对照组、模型组、真武汤组。模型组、真武汤组予阿霉素腹腔注射10周,真武汤组予中药灌胃4周。应用LC-MS技术对大鼠心肌组织进行研究,应用最小判别二乘法,t检验分析代谢物,应用cytoscape关联代谢物-代谢通路,应用Metoboanalyst 5.0对通路进行富集分析。结果模型组有165种差异性表达代谢物。真武汤组有177种差异性表达代谢物,66种为共有。氨基酸生物合成通路、辅助因子合成通路等为相关通路。结论真武汤可通过提高瓜氨酸、丝氨酸等含量,回调多种代谢物含量,发挥抗阿霉素心肌毒性作用,其疗效可能与氨基酸生物合成通路等通路相关。
