摘要
研究青蒿素衍生物KPC-L006的体外抗肺癌活性。以不同浓度KPC-L006处理正常培养的肺癌PC9细胞。采用MTS方法检测KPC-L006处理后对PC9细胞活力的影响;进行细胞划痕实验评价药物对细胞迁移的抑制作用;AnnexinV/PI实验检测细胞凋亡情况;蛋白免疫印迹技术检测凋亡相关蛋白水平的变化。KPC-L006处理后,PC9细胞活力明显下降;划痕修复率降低,细胞迁移能力减弱;细胞凋亡率明显升高;促凋亡相关蛋白Caspase-3,Bax水平提高,抗凋亡蛋白Bcl-2水平降低。KPC-L006具有明显的抗肺癌活性。
Focusing on the investigation of the anti-lung cancer activity of artemisinin derivative KPC-L006 in vitro, Lung cancer PC9 Cell line was processed with KPC-L006 at certain doses. MTS assay was employed to detect the cell viability of PC9 after the treatment with KPC-L006 at following times. Then, scratch cell test was used to assess the inhibitory effect on cell migration from KPC-L006. Cell apoptosis on AnnexinV/PI assay was observed. Measurement of the expression level of apoptosis-related protein was processed by western blotting. After the treatment of KPC-L006, the activity of PC9 cell line decreased significantly. The scratch repair rate was significantly lower. The apoptosis rate increased significantly. The pro-apoptotic protein Cleaved-Caspase 3 and Bax was increased, while the anti-apoptosis protein Bcl-2 was decreased. KPC-L006 had significant anti-lung cancer activity.
