不同产地杠板归总黄酮的含量测定

Determination of Content of Total Flavonoids in Polygonum perfoliatum from Different Areas

为建立杠板归总黄酮的含量测定方法,考察不同产地杠板归药材总黄酮的含量水平,采用分光光度法,以芦丁为对照品,以AlCl_(3)-NaAc溶液为显色剂,检测波长为412 nm检测杠板归总黄酮含量,采用单因素法,分别研究提取溶剂体积分数、固液比、提取时间对总黄酮提取率的影响。芦丁对照品在2.55~25.5μg/mL范围内线性良好(r=0.9988),回归方程为Y=0.03C+0.0065。杠板归总黄酮平均加样回收率为101.7%,RSD为2.3%。10批不同产地杠板归总黄酮含量在0.59%~2.19%之间。该方法简便、稳定,可用于杠板归药材总黄酮的含量测定。

A method for determination of the total flavonoids in Polygonum perfoliatum was established, and the amount of total flavonoids in Polygonum perfoliatum from different production areas was compared. The total flavonoids were determined by UV-Vis spectrophotometer with rutin as reference substance and AlCl_(3)-NaAc as chromogenic agent, the detection wavelength was 412 nm. According to the single factor method, the effects of extraction solvent volume fraction, the ratio of solid to liquid and extraction time were studied. A good linear relationship was achieved over the concentration range of 2.55~25.5 μg/mL(r=0.9988)for rutin, the regression equation was Y=0.03 C+0.0065. The total flavonoid recovery rate of Polygonum perfoliatum was 101.7%,RSD was 2.3%. The content of total flavonoids from 10 different production areas was between 0.59% and 2.19%. The method is simple and stable, and it is suitable for the content determination of total flavonoids of Polygonum perfoliatum.
