摘要
以新鲜苦瓜为原料,洗净低温烘干后,用乙醇水溶液提取,得到苦瓜甙。以得到的苦瓜甙质量为指标,探讨了提取温度、乙醇浓度和提取时间对苦瓜甙得率的影响。在单因素试验基础上,用Box-Behnken中心组合原理建立数学模型,采用响应面法优化得出最佳工艺条件。结果表明,30 g苦瓜干在温度63 ℃、乙醇浓度65%、提取时间3.3 h条件下,得到的苦瓜甙质量最高,为0.7658 g,苦瓜甙的得率为2.553%。同时,HPLC检测结果表明苦瓜提取物中含有苦瓜甙。结果显示,采用响应面法可以较好地优化苦瓜甙的提取工艺,为从苦瓜中提取苦瓜甙的工业化生产提供技术支撑,为苦瓜的深度利用提供了理论依据和优化工艺参数。
Fresh Momordica charantia was washed and dried at low temperature and extracted with ethanol solution to obtain charantin.Taking the quality of charantin as the index,the effects of extraction temperature,ethanol concentration and extraction time on the yield of charantin were studied.On the basis of single factor experiment,a mathematical model was established by using Box-Behnken central combination principle,and the optimal technological conditions were obtained by response surface method.The results showed that the highest quality of charantin was 0.7658 g,and the yield of charantin was 2.553%under the conditions of 63℃,65%ethanol concentration and 3.3 h extraction time.At the same time,the results of HPLC showed that Momordica charantia extract contained charantin.The result further indicated that the response surface method could optimize the extraction process of charantin and provide a technical support for industrial production of charantin and a theoretical basis for deep utilization of Momordica charantia and optimization of technological parameters.
