血清软骨寡聚基质蛋白结合硫酸软骨素846参数对大鼠骨关节炎模型的诊断价值 被引量:1

Diagnostic Value of Combined Serum Cartilage Oligomeric Matrix Protein and Chondroit in Sulfate 846 Parameters in Rat Osteoarthritis Model

摘要 为了全面评估血清中软骨寡聚基质蛋白(COMP)和硫酸软骨素846(CS846)在大鼠骨关节炎(OA)模型中的诊断价值。本试验将60只雄性SD大鼠随机分为对照组(n=30)和模型组(n=30)。其中,模型组通过手术切断前十字韧带(ACLT)造模,对照组进行假手术。造模后第0、2、4、6、8周和第10周,收集血清和胫骨样本,用酶联免疫吸附试验(ELISA)检测各时间点血清中COMP和CS846水平;对大鼠胫骨进行病理组织学检查和国际骨关节炎研究学会(OARSI)评分;用受试者工作特征曲线(ROC)比较单独和结合2个参数检测的曲线下面积(AUC)。结果显示,造模10周内,模型组血清中COMP和CS846水平明显高于对照组(P<0.05),并且COMP和CS846分别与软骨损伤程度呈正相关(P<0.01);结合2个参数检测的AUC值高于单独检测COMP或CS846。因此,结合血清COMP和CS486参数对评估和诊断OA具有潜在的价值。 In order to comprehensively evaluate the diagnostic value of serum cartilage oligomeric matrix protein(COMP) and CS846 in rat osteoarthritis(OA) model, this study randomly divided 60 male SD rats into control group(n=30) and model group(n=30), with the model group being established through anterior cruciate ligament transection(ACLT) surgery, and the control group receiving sham surgery. Serum and tibia samples were collected at 0,2,4,6,8 and 10 weeks after modeling, and the levels of COMP and CS846 in sera were detected by ELISA at each time point. Histopathological examination and Osteoarthritis Research Society International(OARSI) score were performed on rat tibia. Receive operating characteristic curve(ROC curve) was used to compare the Area under curve(AUC) values of the two parameters separately or in combination. The results showed that the serum levels of COMP and CS846 in the model group were significantly higher than those in the control group within 10 weeks after modeling(P<0.05). COMP and CS846 were positively correlated with the degree of cartilage damage(P<0.01). The AUC value of the combination of the two parameters was higher than that of the COMP or CS846 alone. Thus, the combined of serum COMP and CS486 parameters may have potential for improving the evaluation and diagnosis of rat osteoarthritis.
