青海省鼠疫主要宿主寄生蚤携带立克次体调查 被引量:1

Investigation on Rickettsia carried by parasitic fleas in main host of plague in Qinghai Province

摘要 目的对青海省鼠疫主要宿主寄生蚤进行检测,观察立克次体感染情况。方法应用杀虫气雾剂喷杀兴海县、贵南县、玉树州捕获的鼠疫主要宿主喜马拉雅旱獭和青海田鼠,用检蚤镊采集其体表寄生蚤,经鉴定为4科5属6种共120匹。分别对采集的蚤样品单只提取DNA,采用实时荧光定量PCR法(real-time fluorescence quantitative PCR,RT-PCR)检测体表寄生蚤携带的立克次体,并将立克次体反应产物进行琼脂糖凝胶电泳。结果120匹(♂50,♀70)体表寄生蚤中检出4匹立克次体阳性,扩增出目标带,证实有立克次体感染。结论首次自青海省鼠疫主要宿主体表寄生蚤检出立克次体病原,对鼠疫宿主检测及相关传染病的研究提供了基础数据。 Objective To detect main parasitic fleas of plague in Qinghai Province and to observe the infection of Rickettsia.Methods Marmota himalayana and Microtus Qinghai,main hosts of plague,were sprayed with insecticidal aerosol in Xinghai County,Guinan County,and Yushu Prefecture.A total of 120 fleas collected from the body surface of Marmota himalayana and Microtus Qinghai were identified as 6 species,5 genera,4 families.Only DNA was extracted from each flea sample,the Rickettsia carried by the fleas were detected by real-time fluorescence quantitative PCR,and the Rickettsia reaction products were tested by agarose gel electrophoresis.Results Four positive cases of Rickettsia were detected in 120 fleas(♂50,♀70),target bands were amplified,and Rickettsia infection was validated.Conclusion Rickettsia is detected from the parasitic fleas on the surface of main hosts of plague in Qinghai for the first time,providing basic data for the surveillance and detection of main hosts of plague and related infectious diseases.
