电针对慢性应激抑郁大鼠犬尿氨酸代谢途径的影响

摘要 目的观察慢性应激抑郁模型大鼠海马区犬尿喹啉酸(KYNA)、喹啉酸(QUIN)、犬尿氨酸-3-单加氧酶(KMO)、3-羟基邻氨基苯甲酸3,4-双加氧酶(3-HAO)、犬尿氨酸转氧酶(KAT)的变化及电针的干预作用,探讨电针抗抑郁的作用机制。方法按照随机数字表法将24只SD大鼠分成空白组、模型组、氟西汀组、电针组,每组6只。空白组不进行任何干预,其他3组通过慢性温和不可预知性应激刺激结合孤养的方法复制慢性应激抑郁模型;采用开野试验观察大鼠的行为学变化;采用酶联免疫吸附试验(ELISA)检测各组大鼠海马区KYNA、QUIN的变化;实时荧光定量聚合酶链反应(qRT-PCR)检测各组大鼠海马区KMO mRNA、3-HAO mRNA、KAT mRNA相对表达量。结果大鼠开野试验中,模型组大鼠应激前的水平运动次数、垂直运动次数与应激后比较,差异有统计学意义(P<0.05),应激21 d后模型组大鼠水平运动次数、垂直运动的次数较应激前减少,而氟西汀组和电针组可逆转此变化。模型组大鼠海马KYNA含量较空白组降低(P<0.05),电针组大鼠海马KYNA含量较模型组升高(P<0.05);模型组大鼠海马QUIN含量较空白组升高(P<0.05),氟西汀组和电针组大鼠海马QUIN含量较模型组降低(P<0.05)。模型组大鼠海马KMO mRNA、3-HAO mRNA、KAT mRNA相对表达量较空白组升高(P<0.05),氟西丁组和电针组较模型组降低(P<0.05)。结论电针可能通过调节犬尿氨酸代谢途径的异常来达到抗抑郁作用。 Objective To observe the changes of KYNA,QUIN,KMO,3-HAO,KAT in hippocampus of rats with depression,and to reveal the biological mechanisms of electro-acupuncture treating depression.Method Totally 24 SD rats were randomly divided into 4 groups(blank group,model group,fluoxetine group,and electroacupuncture group).The blank group did not receive any intervention,and the other three groups replicated the model of chronic stress depression by chronic mild unpredictable stress stimulation combined with orphanage.The locomotor activity of rats in every group by open-field experiment was observed.Hippocampus were collected,and the level of KYNA,QUIN was detected with Elisa.The KMO mRNA,3-HAO mRNA,and KAT mRNA were detected with real-time PCR.Result In the open field test of rats,there was significant difference in the number of horizontal and vertical movements of rats in the model group before and after stress(P<0.05).After 21 days of stress,the number of horizontal and vertical movements of rats in the model group decreased compared with that before stress,while this change was reversible in fluoxetine group and electroacupuncture group.The content of KYNA in hippocampus of model group was lower than that of blank group(P<0.05),and the content of KYNA in hippocampus of electroacupuncture group was higher than that of model group(P<0.05);The content of QUIN in hippocampus of model group was higher than that of blank group(P<0.05),and the content of QUIN in hippocampus of fluoxetine group and electroacupuncture group was lower than that of model group(P<0.05).The relative expressions of kmo mRNA,3-hao mRNA and KatⅡmRNA in hippocampus of model group were higher than those in blank group(P<0.05),while those in fluoxetine group and electroacupuncture group were lower than those in model group(P<0.05).Conclusion Electroacupuncture may relieve depression through adjusting the abnormal of kynurenine pathway.
