超高压法提取绣球菌多糖工艺优化研究 被引量:2

摘要 目的 优化超高压设备提取绣球菌中多糖的工艺研究。方法 通过单因素试验考察提取压力、保压时间、料液比、提取液pH和提取温度对绣球菌多糖得率的影响。在单因素试验的基础上,通过Plackett-Burman设计筛选出影响显著因素,并对显著因素进行最陡爬坡试验,使显著因素的取值逼近最大响应区域,确定Box-Behnken试验的中心点,最后采用Box-Behnken试验对绣球菌多糖得率影响显著的3个因素进行预测,指导提取工艺条件优化,并通过试验进行验证。结果 在保压时间为15min、料液比为1:40(g/mL)条件下,绣球菌多糖的优化提取工艺为:提取压力315 MPa,提取液pH 9.2,提取温度48.5℃。在此工艺条件下,测得绣球菌多糖的得率为14.72%±0.15%,与预测值14.83%接近,说明模型准确。结论 本研究建立的绣球菌多糖提取工艺切实可行,可用于绣球菌多糖的规模化提取。 Objective To optimize the extraction process of polysaccharide from Sparassis crispa by ultra high pressure equipment. Methods Conducting single factor experiment to investigate the effects of extraction pressure,pressure holding time, solid-liquid ratio, pH of extraction solution and extraction temperature on the yield of Sparassis crispa polysaccharide. On the basis of the single factor experiment, Plackett-Burman design was used to screen out the significant factors, and the steepest climbing experiment was carried out for the significant factors so that the value of the significant factors was close to the maximum response region, then the center point of the Box-Behnken experiment was determined. Finally, Box-Behnken experiment was used to predict the three factors that had significant influence on the extraction rate of Sparassis crispa polysaccharide, to guide the optimization of extraction conditions, and to verify the experiment. Results Under the premise of fixed pressure holding time of 15min and solid-liquid ratio of 1:40(g/mL), the optimum extraction process of Sparassis crispa polysaccharide was as follows: Extraction pressure 315 MPa, pH 9.2 and extraction temperature 48.5 ℃. Under these conditions, the yield of Sparassis crispa polysaccharide was 14.72%±0.15%, which was close to the predicted value of 14.83%, indicating that the model was accurate. Conclusion The extraction process of Sparassis crispa polysaccharide established in this study is feasible and can be used for the extraction of Sparassis crispa polysaccharide.
