新型蓝莓花色苷含量测定及抗氧化性能研究 被引量:7

摘要 为探究我国自主研发的国家级林木良种“蓝美1号”提取物蓝莓粉(blueberry No.1 podwer,B1P)的功能成分及体外抗氧化能力,采用pH示差法测定B1P总花色苷含量,利用超高效液相串联质谱(ultra performance liquid chromatography-mass spectrum,UPLC-MS/MS)测定其花色苷种类及含量,同时以V_(C)为对照品考察不同浓度B1P的体外抗氧化能力。结果表明:B1P总花色苷含量为(443.08±16.40)mg/g;建立一种能够同时检测8种花色苷的UPLC-MS/MS方法,在0.5 ng/mL~50.0 ng/mL浓度范围内线性良好,相关系数R^(2)均大于0.9900。测得的8种花色苷中含量最高为飞燕草素葡萄糖苷(80.90±0.30)mg/g,最低为芍药素葡萄糖苷(6.03±0.02)mg/g。在10μg/mL~100μg/mL内,B1P对ABTS^(+)·、羟基自由基、超氧阴离子自由基的清除能力均优于阳性对照V_(C)(p<0.05),且对ABTS^(+)·、羟基自由基、超氧阴离子自由基半数抑制浓度(IC_(50))分别为(37.81±4.30)、(51.33±8.03)、(19.88±2.88)μg/mL。此蓝莓中含大量以飞燕草素和矮牵牛素为苷元的糖苷,多种花色苷协同清除自由基,抗氧化效果良好。 In order to explore the functional components and in vitro antioxidant capacity of blueberry No.1 powder(B1P),an extract of“Lanmei No.1”was independently developed by China.The total anthocyanin content of B1P was determined by the pH differential method,and the anthocyanin types and contents were determined by ultra-high performance liquid chromatography-tandem mass spectrum(UPLC-MS/MS).Simultaneously,the antioxidant capacity of B1P at different concentrations in vitro was investigated with V_(C) as the reference.The results showed that the total anthocyanin content of B1P was(443.08±16.40)mg/g.An UPLC-MS/MS method for simultaneous detection of eight anthocyanins,was established.The linearity,which was in the concentration range of 0.5 ng/mL to 50.0 ng/mL,was good,and the correlation coefficients R^(2) were all greater than 0.9900.Among the eight anthocyanins,the highest anthocyanin content was exhibited by delphinidin glucoside(80.90±0.30)mg/g,and the lowest anthocyanin content was exhibited by peonidin glucoside(6.03±0.02)mg/g.In the experimental concentration range of 10μg/mL-100μg/mL,the scavenging abilities of ABTS^(+)·,·OH,and superoxide anion were better than that of positive control V_(C)(p<0.05),and the IC_(50) values of B1P to ABTS^(+)·,·OH,and superoxide anion were(37.81±4.30)μg/mL,(51.33±8.03)μg/mL and(19.88±2.88)μg/mL,respectively.Thus,it was concluded that this blueberry contains a large amount of glucosides with delphinidin and peonidin as aglycones.Further,various anthocyanins co-operate to scavenge free radicals with good antioxidant effect.
