TGM2、NEAT1、miRNA-17-5p在胃癌组织中的表达及与疾病演进的相关性 被引量:4

Expressions of TGM2,NEAT1 and miRNA 175p in gastric cancer tissue and their correlation with disease progression

摘要 目的研究转谷氨酰胺酶2(transglutaminase,TGM2)、核富集转录本1(nuclear paraspeckle assembly transcript1,NEAT1)、miRNA-17-5p在胃癌(gastric cancer,GC)组织中的表达及与疾病演进的相关性。方法选取病理科保存的GC组织标本74例,为研究组,另选取同一时间段病理科保存的正常胃黏膜(距离癌组织>5 cm)74例,为对照组,检测TGM2、NEAT1、miRNA-17-5p表达水平,统计研究对象一般资料及GC患者与疾病演进的相关性。结果与对照组比较,研究组TGM2、NEAT1阳性表达率较高,miRNA-17-5p阳性表达率较低,差异有统计学意义(P<0.05)。与无淋巴转移比较,有淋巴转移患者TGM2、NEAT1阳性表达率较高,miRNA-17-5p阳性表达率较低;与临床Ⅲ~Ⅳ期比较,Ⅰ~Ⅱ期患者TGM2、NEAT1阳性表达率较高,miRNA-17-5p阳性表达率较低;与黏膜下层比较,TGM2、NEAT1阳性表达率较高,miRNA-17-5p阳性表达率较低,差异有统计学意义(P<0.05)。Logistic回归分析TGM2、NEAT1、miRNA-17-5p阳性表达率升高与发生GC独立相关(P<0.05)。受试者工作特征曲线(ROC)分析TGM2、NEAT1、miRNA-17-5p预测发生GC曲线下面积(AUC)分别为0.662、0.716、0.754。结论GC患者组织中TGM2、NEAT1阳性表达率增高,miRNA-17-5p阳性表达率降低,其阳性表达的高低与患者淋巴转移、临床分期、病灶深度显著相关,在临床诊断中具有重要意义。 Objective To investigate the expressions of transglutaminase(TGM2),nuclear paraspeckle assembly transcript1(NEAT1)and miRNA-17-5p in gastric cancer(GC)tissue and their correlation with disease progression.Methods A total of 74 cases of GC tissues which were preserved in pathology department of our hospital from December 2018 to November 2020 were enrolled as research group,at the same time,the other 74 cases of normal GC tissues(more than 5cm from cancer tissue)were enrolled as control group.The expression levels of TGM2,NEAT1 and miRNA-17-5p were detected,and the general data of patients,and the correlation between GC patients and disease progression were statistically analyzed.Results The positive expression rates of TGM2,NEAT1 in research group were significantly higher than those in control group,however the miRNA-17-5p positive expression rate was significantly lower than that in control group(P<0.05).As compared with those of patients without lymphatic metastasis,the positive expression rates of TGM2,NEAT1 in patients with lymphatic metastasis were significantly increased,however the miRNA-17-5p positive expression rate was significantly decreased(P<0.05).As compared with those in patients of clinicⅢ~Ⅳstage,the positive expression rates of TGM2,NEAT1 in patients of clinicⅠ~Ⅱstage were significantly increased,however the miRNA-17-5p positive expression rate was significantly decreased(P<0.05).As compared with those of cells in submucosa,the positive expression rates of TGM2,NEAT1 of cells in gastric mucosa were significantly increased,however the miRNA-17-5p positive expression rate was significantly decreased(P<0.05).Logistic regression analysis showed that the increaseed positive expression rates of TGM2,NEAT,miRNA-17-5p were independently related to the occurrence of GC(P<0.05).The subject job characteristic curve(ROC)analysis showed that area under the curve(AUC)of TGM2,NEAT1 and miRNA-17-5p were 0.662,0.716,0.754,respectively.Conclusion The positive expression rates of TGM2 and NEAT1 are increased in patients with GC,however the positive expression rate of miRNA-17-5p is decreased.Moreover the positive expression levels of TGM2 and NEAT1 are closely correlated with lymph node metastasis,clinical stage and disease focus depth,which are of important significance in clinical diagnosis.
