摘要
目的证实母代孕期和哺乳期高脂饮食可敏化雄性子代的血压中枢,使雄性子代增加了血管紧张素Ⅱ(angitensinⅡ,AngⅡ)的敏感性,导致其成年后暴露在高盐饮食下,更易发生高血压。方法正常成年SD雌鼠随机分为2组,与健康雄鼠交配前2周,分别予正常饮食(C)和高脂饮食(H)喂养,直至子代出生后3周断乳为止,将2组内雄性子代随机分为正常饮食(C/MC和H/MC)和高盐饮食(C/MH和H/MH)组喂养,子代10周龄时外周注射苯肾上腺素/硝普钠观察压力感受器反射敏感性变化;全身慢性灌注或急性侧脑室微量注射AngⅡ比较各组血压、心率反应性。结果高脂饮食组的高盐饮食子代对慢性灌流AngⅡ的升压效应明显加强,对急性侧脑室注射AngⅡ后的升压效应更明显。结论母代高脂饮食可上调雄性子代中枢升压核团反应性和降低压力感受器反射敏感性,导致其在成年后暴露在高盐饮食环境下,更易发生高血压。
Objective To investigate the effects of Effects of high-fat diet on the blood pressure of male offspring of maternal sensitization through injection of angiotensinⅡ(AngⅡ).Methods The adult SD female rats were randomly divided into two groups randomly,who were fed with the normal diet(control group)and high fat diet(observation group)at two weeks before mating with healthy fema rats.Moreover the offsprings of the two groups were randomly divided into normal salt diet(C/MC and H/MC)geoup and high salt diet(C/MH and H/MH)group.The sensitivity of baroreflex were assessed through peripheral injection of phenylephrine/sodium nitroprusside at the age of 10 weeks.The blood pressure and heart rate responses were observed by systemic acute or chronic perfusion of AngⅡ.Results The levels of blood pressure of offspring in high fat diet group were significantly increased,which was more obvious after lateral ventricle acute perfusion.Conclusion Maternal high fat diet can increase the brain reactivity and blunted baroreflex in male offsprings fed with high salt diet,thereby leading to susceptibility to develop hypertension.
