经口灌胃熊果酸对小鼠结肠、肝和肾组织抗氧化能力的影响 被引量:3

Effect of Intragastric Administration of Ursolic Acid on Antioxidant Capacities of Colon,Liver and Kidney in Mice

摘要 本试验旨在探讨熊果酸经口灌胃对ICR小鼠结肠、肝和肾组织抗氧化能力的影响。将50只ICR小鼠随机分为空白对照组、溶剂对照组、熊果酸低、中和高剂量组[5、10 mg/(kg·bw)和15 mg/(kg·bw)]。熊果酸低、中和高剂量组给予熊果酸连续灌胃28 d,溶剂对照组给予相同剂量的0.5%羧甲基纤维素钠溶液灌胃。摘取结肠、肝和肾组织,分光光度法检测组织抗氧化酶活力[(过氧化物酶(POD)、过氧化氢酶(CAT)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、一氧化氮合酶(NOS)、总抗氧化能力(T-AOC)和脂质过氧化物(LPO)]和氧化损伤标志物水平[(一氧化氮(NO)、过氧化氢(H_(2)O_(2))和丙二醛(MDA)]。结果显示:口服熊果酸可以降低小鼠结肠组织的抗氧化酶活力(POD、CAT、SOD、GSH-Px、T-AOC和LPO),升高氧化损伤标志物的水平(MDA、H_(2)O_(2)和NO);小鼠肝组织和肾组织的抗氧化酶活力(GSH-Px、CAT、POD和SOD)显著提高。鉴于熊果酸具有可逆性地引起小鼠结肠组织抗氧化能力下降的能力,其可能起到保护肝肾组织的作用。 This study aimed to investigate the antioxidant capacity of ursolic acid on the colon,liver and kidney of ICR mice through continuous intragastric administration.50 ICR mice were randomly divided into blank control group,solvent control group,low-,medium-,and high-dose group of ursolic acid[5,10 mg/(kg·bw)and 15 mg/(kg·bw)].Ursolic acid solution was given to ursolic acid groups by intragastric administration for 28 days,while the same dose of 0.5%carboxymethyl cellulose was given to solvent control group.Mice were sacrificed to collect colon,liver,and kidney tissues for assessing the antioxidant enzymes[peroxidase(POD),catalase(CAT),superoxide dismutase(SOD),glutathioneperoxidase(GSH-Px),nitric oxide synthase(NOS),total antioxidant capacity(T-AOC),and lipid peroxidation(LPO)]and oxidative damage markers[nitric oxide(NO),hydrogen peroxide(H_(2)O_(2)),and malondialdehyde(MDA)]by spectrophotometric method.The results showed that the activities of antioxidant enzymes(POD,CAT,SOD,GSH-Px,T-AOC,and LPO)in colon tissues were reduced by ursolic acid,whereas the levels of oxidative damage markers(MDA,H_(2)O_(2),and NO)were increased.Further,the activities of antioxidant enzyme(GSH-Px,CAT,POD,and SOD)in liver and kidney tissues were also increased by ursolic acid.Given its ability to reversibly reduce the antioxidant capacity of colon tissues in mice,ursolic acid may have protective effects on liver and kidney tissues.
