狭叶重楼中一个新的环胆甾烷醇型甾体皂苷 被引量:2

摘要 目的研究狭叶重楼的皂苷类化学成分,为拓宽重楼属植物的药用资源提供科学依据。方法采用硅胶柱色谱、Sephadex LH-20凝胶柱色谱、反相硅胶柱色谱和半制备高效液相色谱等方法,从狭叶重楼70%乙醇提取物的乙酸乙酯萃取部位和正丁醇萃取部位分离化合物,通过波谱分析和化学手段鉴定化学结构。结果分离得到7个甾体皂苷类成分,分别鉴定为26-O-β-D-吡喃葡萄糖基-3β,16α,26-三羟基-(23R,25R)-16,23-环胆甾烷醇-5,17(20)-二烯-22-酮-3-O-α-L-呋喃阿拉伯糖基-(1→4)-[α-L-吡喃鼠李糖基-(1→2)]-β-D-吡喃葡萄糖苷(1)、重楼皂苷H(2)、重楼皂苷Ⅵ(3)、偏诺皂苷元-3-O-α-L-吡喃鼠李糖基-(1→4)-[α-L-吡喃鼠李糖基-(1→2)]-β-D-吡喃葡萄糖苷(4)、重楼皂苷Ⅶ(5)、26-O-β-D-吡喃葡萄糖基-17,22-二羟基-(25R)-呋甾-5-烯-3-O-α-L-呋喃阿拉伯糖基-(1→4)-[α-L-吡喃鼠李糖基-(1→2)]-β-D-吡喃葡萄糖苷(6)和26-O-β-D-吡喃葡萄糖基-17,22-二羟基-(25R)-呋甾-5-烯-3-O-α-L-吡喃鼠李糖基-(1→4)-吡喃鼠李糖基-(1→4)-[α-L-吡喃鼠李糖基-(1→2)]-β-D-吡喃葡萄糖苷(7)。结论化合物1为新化合物,是具有罕见骨架结构的环胆甾烷醇型甾体皂苷,为首次从重楼属植物中发现,化合物4、6和7为首次从狭叶重楼中分离得到。 Objective To determine the saponin constituents from the rhizomes of Paris polyphylla var.stenophylla to enrich the resource of the plants from genus Paris.Methods The compounds from the ethyl acetate fraction and n-butanol fraction of 70%ethanol extraction were isolated and purified by silica gel column chromatography,Sephadex LH-20 column chromatography,ODS column chromatography and semi-preparative HPLC.Their structures were identified by spectral analysis and chemical methods.Results Seven steroidal saponins were isolated and identified as 26-O-β-D-glucopyranosyl-3β,16α,26-trihydroxy-(23R,25R)-16,23-cyclocholest-5,17(20)-dien-22-one-3-O-α-L-arabinofuranosyl-(1→4)-[α-L-rhamnopyranosyl-(1→2)]-β-D-glucopyranoside(1),paris saponin H(2),polyphyllinⅥ(3),pennogenin-3-O-α-L-rhamnopyranosyl-(1→4)-[α-L-rhamnopyranosyl-(1→2)]-β-D-glucopyranoside(4),polyphyllinⅦ(5),26-O-β-Dglucopyranosyl-17,22-dihydroxy-(25R)-furost-5-en-3-O-α-L-arabinofuranosyl-(1→4)-[α-L-rham-nopyranosyl-(1→2)]-β-D-glucopyranoside(6)and 26-O-β-D-glucopyranosyl-17,22-dihydroxy-(25R)-furost-5-en-3-O-α-L-rhamnopyranosyl-(1→4)-O-α-L-rhamnopyranosyl-(1→4)-O-[α-L-rha-mnopyranosyl-(1→2)]-β-D-glucopyranoside(7),respectively.Conclusion Compound 1 is a new cyclocholestanol-type steroidal saponin with a rare skeleton,which has been firstly found from genus Paris.Compounds 4,6 and 7 have been isolated from the plant for the first time.
