摘要
目的首次研究具柄重楼根茎的甾体皂苷类成分。方法采用硅胶柱色谱、反相ODS柱色谱、Sephadex LH-20凝胶柱色谱和半制备高效液相色谱等方法,从具柄重楼根茎70%乙醇提取物的正丁醇萃取部位分离化合物,通过波谱分析和化学手段鉴定化学结构。结果分离得到7个甾体皂苷类成分,分别鉴定为protogracillin(1)、dichotomin(2)、parisyunnanoside A(3)、saponin Th(4)、26-O-β-D-吡喃葡萄糖基-3β,26-二羟基-(25R)-胆甾-5,17(20)-二烯-16,22-二酮-3-O-α-L-呋喃阿拉伯糖基-(1→4)-[α-L-吡喃鼠李糖基-(1→2)]-β-D-吡喃葡萄糖苷(5)、parispseudoside C(6)和ypsiyunnoside A(7)。结论化合物1~7均为首次从具柄重楼中分离得到,化合物5~7为重楼属植物中少见的胆甾烷醇型甾体皂苷,且化合物7的苷元具有罕见的骨架结构,为首次在重楼属植物中发现。
Objective To determine the saponin constituents in the rhizomes of Paris fargesii var.petiolate for the first time.Methods Compounds from the n-butanol fraction of 70%ethanol extraction were isolated and purified by column chromatographies of silica gel,ODS,Sephadex LH-20 and semi-preparative HPLC,and their structures were identified by spectroscopic analysis and chemical methods.Results Seven steroidal saponins were isolated and identified as protogracillin(1),dichotomin(2),parisyunnanoside A(3),saponin Th(4),26-O-β-D-glucopyranosyl-3β,26-dihydroxy-(25R)-cholest-5,17(20)-dien-16,22-dione-3-O-α-L-arabinofuranosyl-(1→4)-[α-L-rhamnopyranosyl-(1→2)]-β-D-glucopyranoside(5),parispseudoside C(6)and ypsiyunnoside A(7),respectively.Conclusion Compounds 1~7 have been obtained from Paris fargesii var.petiolate for the first time.Compounds 5~7 are cholesteranol-type saponinsrarely seen in genus Paris,and the aglycone of compound 7 possesses a rare skeleton firstly found in the genus.
