基于LC-MS法分析宁夏贺兰山野生酸枣不同部位化学成分 被引量:5

摘要 目的基于高分辨色谱与质谱联用分析平台,对贺兰山野生酸枣不同部位的化学成分进行定性分析,促进野生酸枣资源的开发和利用。方法以50%甲醇对酸枣的果仁、叶片、果肉进行提取,色谱分析使用Waters ACQUITY UPLC BEH C_(18)色谱柱,流动相为0.1%甲酸水(A)-0.1%甲酸乙腈(B)。采用UPLC-Q-TOF/MS^(E)和UHPLC-Q-Exactive/MS技术对酸枣各部位中的化合物进行鉴定。数据采集分别采用MassLynx和Xcalibur工作站对一级碎片进行分析,并结合UNIFI软件及文献信息对二级碎片进行鉴定。结果鉴定出贺兰山野生酸枣中共有62种化合物,其中黄酮类化合物20种,生物碱类化合物8种,萜类化合物12种,脂肪酸类化合物8种,核苷类化合物5种,氨基酸类化合物5种,有机酸类化合物3种,甾体类化合物1种。结论黄酮类及萜类化合物主要分布在果仁和叶片中,生物碱类和脂肪酸类化合物在果仁、叶片、果肉位分布较均匀,通过探究野生酸枣不同部位的成分差异,为贺兰山野生酸枣资源的开发利用提供一定的理论基础。 Objective To qualitatively analyze chemical constituents from different parts of wild sour jujube in Helan Mountains by high-performance liquid chromatographic tandem mass spectrometry and to promote the development and utilization of wild sour jujube resources.Methods The kernels,leaves and sarcocarp of wild sour jujube were extracted with 50%methanol,and the separation was performed on Waters ACQUITY UPLC BEH C_(18) column.The mobile phase was 0.1%formic acid(A)-0.1%formic acid acetonitrile(B)for the gradient elution.UPLC-Q-TOF/MS^(E) and UHPLC-QExactive/MS techniques were used to identify the compounds in each part of the wild sour jujube.Data acquisition was performed with MassLynx and Xcalibur work stations for analysis of primary fragments.UNIFI software and literature information were used to identify the secondary fragments.Results Totally 62 compounds were identified,including 20 flavonoids,8 alkaloids,12 terpenoids,8 fatty acids,5 nucleosides,5 amino acids,3 organic acids and 1 steroidal compound.Conclusion Flavonoids and terpenoids are mainly distributed in the kernels and leaves,while alkaloids and fatty acids are more evenly distributed in the kernels,leaves and sarcocarp.By exploring the differences in the compositions from different parts of wild sour jujube,we provide theoretical basis for the development and utilization of wild sour jujube resources in Helan Mountains.
