赶黄草活性成分槲皮素和没食子酸对CCl_(4)所致小鼠急性肝损伤的保护作用及机制 被引量:9

Protective effects and mechanisms of quercetin and gallic acid from active constituents of Penthorum on acute liver injury induced by CCl_(4) in mice

摘要 目的:探究赶黄草活性成分槲皮素和没食子酸对CCl_(4)所致小鼠急性肝损伤的保护作用及机制。方法:将小鼠随机分为4组(每组10只):正常组、模型组、槲皮素组(100 mg/kg)和没食子酸组(100 mg/kg)。正常组和模型组小鼠灌胃等量生理盐水,其余各组按分组给予相应药物灌胃7 d,末次灌胃后2 h,腹腔注射0.2%CCl_(4)植物油溶液(15 ml/kg),并16 h后收集小鼠血清和肝脏组织。HE染色观察小鼠肝脏病理变化;ELISA检测小鼠血清谷氨酸氨基转移酶(ALT)、肿瘤坏死因子-α(TNF-α)和肝组织还原型谷胱甘肽(GSH)水平;Western blot和免疫荧光检测肝脏中血红素加氧酶-1(HO-1)水平。结果:相比于模型组小鼠,槲皮素和没食子酸干预后,可降低其肝脏指数(P<0.05),降低血清ALT和TNF-α水平(P<0.01),增加肝组织中GSH水平(P<0.01),增加肝组织中HO-1表达。结论:赶黄草活性物质槲皮素和没食子酸对于肝脏损伤具有保护作用,其机制可能与增强抗氧化损伤能力,降低炎症因子水平有关。 Objective:To investigate the protective effect and mechanism of quercetin and gallic acid from active constituents of Penthorum Chinese Pursh on acute liver injury induced by CCl_(4) in mice.Methods:Mice were randomly divided into 4 groups(10 in each group): normal control group, model group, quercetin group(100 mg/kg) and gallic acid group(100 mg/kg). Normal group and model group were given equal amount of normal saline, the rest groups were given 7 days at dose, and 0.2% CCl_(4) vegetable oil solution(15 ml/kg) was injected intraperitoneally 2 hours after last administration, and mice serum and liver tissue were collected after 16 hours. The liver pathological changes of mice were observed by HE staining in paraffin sections and the levels of glutamate aminotransferase(ALT),tumor necrosis factor-α(TNF-α) and reduced glutathione(GSH) were detected by ELISA. The level of heme oxygenase-1(HO-1) in liver was detected by Western blot. Results:Compared with the model group, quercetin and gallic acid could decrease liver index(P<0.05),ALT level(P<0.01),TNF-α level(P<0.01) and GSH level in liver tissue(P<0.01),The expression of HO-1 in liver tissue was increased. Conclusion:Both quercetin and gallic acid from active constituents of Penthorum have protective effects on liver injury, the mechanism of which is related to the enhancement of antioxidant damage and the decrease of inflammatory factor level.
