摘要
目的:探讨人脐带间充质干细胞(UC-MSCs)复合丝心蛋白/胶原/聚(左旋乳酸-己内酯)(SF/COL/PLCL)静电纺丝纳米纤维支架构建盆底重建组织工程补片的可行性。方法:培养UC-MSCs,构建慢病毒介导的绿色荧光蛋白标记的UC-MSCs,制备SF/COL/PLCL静电纺丝三维纳米纤维支架并复合形成新型组织工程盆底补片,同时观察形态。将组织工程补片植入相应大鼠皮下,对照组大鼠植入纳米纤维支架,观察其在体细胞相容性、力学性能及体内转归情况。结果:UC-MSCs体外增殖良好,SF/COL/PLCL静电纺丝纳米纤维支架性能良好,UC-MSCs可黏附于支架并形成细胞层。负载UC-MSCs补片在体生物相容性好,负载UC-MSCs补片组的支架周围组织中白细胞介素2(IL-2)、IL-6、IL-8和肿瘤坏死因子α表达水平随着在体时间延长下降速率相对较快,更利于血运重建,杨氏模量优于对照组(P<0.05),胶原含量增长更快,在体转归分布良好。结论:负载UC-MSCs补片展现了良好的细胞相容性,更强的顺应性和韧性,以及良好的抗炎性和组织修复能力,可为盆底重建提供新思路。
AIM:To investigate the feasibility of human umbilical cord mesenchymal stem cells(UC-MSCs) combined with silk fibroin/collagen/poly(L-lactide-co-ε-caprolactone)(SF/COL/PLCL)electrospun nanofiber scaffolds for pelvic floor reconstruction tissue-engineering mesh.METHODS:Lentivirus-mediated green fluorescent protein-labeled UC-MSCs were constructed.The SF/COL/PLCL electrospun three-dimensional nanofiber scaffolds were prepared.The tissue-engineering patches were implanted subcutaneously in the abdomens of rats,while the rats implanted with the nanofiber scaffolds without UC-MSCs served as control group.The biocompatibility,mechanical properties and in vivo fate of the nanofiber scaffolds were observed.RESULTS:The SF/COL/PLCL electrospun nanofiber scaffolds had good performance.The UC-MSCs grew well in vitro and were attached to scaffolds.Compared with control group,the expression levels of interleukin-2(IL-2),IL-6,IL-8 and tumor necrosis factor-αin UC-MSCs mesh group decreased at a faster rate(P<0.05).Meanwhile,the content of collagen was increased and better body distribution were observed.CONCLUSION:The implanted patches seeded with UC-MSCs show better cytocompatibility,stronger compliance and toughness,and better antiinflammatory and tissue repair capabilities than the naked patches,which meet the requirements of pelvic floor reconstruction.
