摘要
阿尔茨海默病(Alzheimer disease,AD)作为一种以进行性认知功能障碍和行为损害为主要临床表现的进行性中枢神经系统(central nervous system,CNS)变性疾病,在老龄化加速的现代社会越来越受到重视[1-2]。AD的主要病理表现为β-淀粉样蛋白(amyloidβ-protein,Aβ)斑块的累积、tau蛋白神经原纤维缠结和系列神经炎症[3]。
Microglia are innate immune cells in the central nervous system,and their phagocytic activity plays an important role in neurodegenerative diseases such as Alzheimer disease(AD).In previous studies,microglia phagocytosis has been simply regarded as a beneficial treatment for AD.To some extent,the phagocytosis of AD pathological products by microglia can delay the disease progression.However,the effect of phagocytosis on microglia themselves is ignored.Excessive phagocytosis will lead to the damage of microglia,decline of phagocytosis,and even cause pro-inflammatory polarization of microglia and increase neurotoxicity.This article reviews the role and mechanism of microglial phagocytosis and discuss the effects of targeted regulation of microglial phagocytosis in different stages of AD,in order to provide some new ideas for the therapeutic strategy of AD.
