甘草养阴汤对老年类风湿关节炎的价值探索 被引量:2

摘要 目的:探索甘草养阴汤(Gancao Nurish-Yin decoction,GCNY)干预类风湿关节炎(Rheumatoid Arthritis,RA)合并衰老的基因靶点及分子机制。方法:通过中药系统药理学数据库及分析平台(Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform,TCMSP)、和中药综合资源数据库(The Encyclopedia of Traditional Chinese Medicine,ETCM)获得甘草养阴汤7种中药的有效成分,及其作用的基因靶点。通过GeneCards数据库获取RA及衰老相关基因靶点,获取二列基因交集,即GCNY干预RA合并衰老的基因靶点。对这些潜在作用靶点进行GO(Gene ontology)、KEGG(Kyoto Encyclopedia of Genes and Genomes)基因功能注释及通路富集分析。结果:GCNY7种中药中总共含有130种口服可吸收的有效成分,共277个靶点基因;RA相关靶点基因共393个、衰老相关靶点基因共216个,两者共同基因靶点61个。GCNY与RA和衰老靶点的交集基因共有26个,包括PTGS2、NOS2、TGFB1、TNF、MMP1、IL1B、STAT3等基因。参与对氧化应激的反应、对细菌来源的分子的反应、对脂多糖的反应、活性氧代谢过程的调节等。结论:GCNY通过调控RA合并衰老的多个基因靶点,可能在老年性RA患者的治疗中发挥优势。 Objective:To explore the gene target and molecular mechanism of Gancao Yangyin decoction(GCNY)intervention in Rheumatoid Arthritis(RA)complicated with aging.Methods:Through the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP)and the Traditional Chinese Medicine Comprehensive Resource Database(The Encyclopedia of Traditional Chinese Medicine,ETCM),the efficacy of seven traditional Chinese medicines of Gancao Yangyin decoction was obtained.components,and their gene targets of action.The RA and aging-related gene targets were obtained through the GeneCards database,and the two-column gene intersection was obtained,that is,the gene targets of GCNY intervention in RA complicated with aging.GO(Gene ontology)and KEGG(Kyoto Encyclopedia of Genes and Genomes)gene function annotation and pathway enrichment analysis were performed on these potential targets.Results:7 kinds of traditional Chinese medicines in GCNY contained a total of 130 orally absorbable active ingredients,with a total of 277 target genes;a total of 393 RA-related target genes,216 aging-related target genes,and 61 common gene targets..There are 26 intersection genes between GCNY and RA and aging targets,including PTGS2,NOS2,TGFB1,TNF,MMP1,IL1B,STAT3 and other genes.Involved in the response to oxidative stress,the response to bacteria-derived molecules,the response to lipopolysaccharide,the regulation of reactive oxygen species metabolic processes,etc.Conclusion:GCNY may play an advantage in the treatment of elderly RA patients by regulating multiple gene targets of RA complicated with aging.
