摘要
本文优化并建立了高效液相色谱法和离子色谱法测定低聚半乳糖原料中不同聚合度低聚半乳糖组分的比值,解决了《离子色谱法检测食品中的低聚半乳糖》(AOAC.2001.02)方法中产品聚合度不确定的问题。本文以配方食品中添加的低聚半乳糖原料为对象,利用高效液相色谱法测定低聚半乳糖原料中二糖、三糖、四糖、五糖、六糖及六糖以上各组分占总糖的比值,再利用离子色谱法分离并测定低聚半乳二糖和乳糖含量,通过计算得到低聚半乳糖原料的平均聚合度。用面积归一化法测定半乳糖和1,3(4)-β-D-半乳二糖。该方法检测结果准确,前处理操作简单,便于检测原料产品中低聚半乳糖原料的聚合度组成。
In this paper,high performance liquid chromatography and ion chromatography were optimized and established to determine the ratio of galactooligosaccharides with different degrees of polymerization in galactooligosaccharides,which solved the problem of uncertainty of degree of polymerization in AOAC.2001.02.In this paper,the ratio of disaccharide,disaccharide,trisaccharide,tetrasaccharide,pentasaccharide,hexasaccharide to total sugar in the raw material of galactooligosaccharide was determined by HPLC method,and the content of galactooligosaccharide and lactose was separated and determined by ion chromatography.The average polymerization degree of the raw material of galactooligosaccharide was obtained by calculation.Determination of Galactose and 1,3(4)-β-D-galactobiose by area normalization method.The results of this method are accurate,and the pretreatment operation is simple.It is easy to detect the polymerization degree of galactooligosaccharide in raw materials.
