基于高通量测序的瞿麦叶片转录组分析

Analysis of the Transcriptome of Dianthus superbus Based on High Throughput Sequencing

摘要 为获得瞿麦(Dianthus superbus L.)的基因数据并了解其基因功能,本研究通过高通量测序技术对瞿麦幼苗叶片的转录组进行测序,并对其进行生物信息学分析。通过Illumina测序平台共获得10825条Unigene,总长度为15384267 bp。将获得的Unigene分别与Nr、GO、KOG等7大数据库进行比对,最终有8255条(76.26%)Unigene获得注释。其中8201条Unigene在Nr数据库中获得注释,注释同源物种主要是甜菜、藜麦;与GO数据库比对后获得注释Unigene 5886条,分为细胞组分、生物进程和分子功能3个功能大类23个亚类;KEGG数据库共获得注释Unigene 6204条,共涉及6大类45个亚类346个代谢通路,其中以新陈代谢相关通路分布较多;KOG数据库共获得注释Unigene 5303条,分为25个功能类别,基因注释最多的为一般功能和翻译后修饰;在所有Unigene中共检测到SSR位点5201个,以单核苷酸和三核苷酸重复为主。本研究为开展瞿麦功能基因挖掘、次生代谢途径解析和遗传多样性分析提供了理论基础。 In order to obtain the genetic data of Dianthus superbus L.and study its gene function,in this study,the transcriptome of D.superbus leaves was sequenced by Illumina high-throughput sequencing technology,and further analyzed by bioinformatics.10825 Unigenes were generated from Illumina sequencing platform,with a total length of 15384267 bp.8255 Unigenes(76.26%)were successfully annotated against 7 public bioinformatics databases.In Nr database,8201 Unigenes were annotated mainly from Beta vulgaris and Chenopodium quinoa;5886 Unigenes were annotated in GO database and were divided into cellular component,biological progress,and molecular functions with 23 subgroups;6204 Unigenes were classified into 346 metabolic pathways through KEGG database,in which metabolism-related pathways were widely distributed;there were 5303 Unigenes annotated in 25 KOG function categories,the most annotated genes were involved in general function prediction;5201 SSR sites were detected in all Unigenes,mainly including Mono-nucleotide and Tri-nucleotide repeats.This study provided scientific evidence for the research of functional gene identification,secondary metabolism pathway disssection and genetic diversity analysis in D.superbus.
