摘要
棉花枯萎病是海岛棉的主要病害,近年来由于连年种植,枯萎病已严重影响海岛棉生产,由于其复杂性有关海岛棉本身枯萎病抗性基因的定位和克隆的研究报道较少。本研究基于前期QTL位点qFw1-1(Fw3),利用侧冀近端紧密连锁分子标记鉴定和组配F2群体接种鉴定获得Fw3位点单基因系,以F2群体抗感单株符合3:1的群体为定位群体,设计引物对隐形群体进行初步定位。结果显示Fw3定位在A01号染色体的标记ZJUd03067与ZJUd03018之间,与这2个标记的遗传距离分别为7.8 cM和10.7 cM,该区间共含有67个基因。本研究为海岛棉枯萎病的抗性基因克隆和分子机理解析提供参考。
Cotton Fusarium wil is the main disease of sea-island cotton.In recent years,due to years of cultivation,Fusarium wilt has seriously affected island cotton production.Because of its complexity,there are few studies on the positioning and cloning of Fusarium wilt resistance genes in sea-island cotton itself.This study is based on the previous QTL site qFw1-1(i.e.Fw3 in this paper).Fw3 site monogeneic lines were obtained by identification of closely linked molecular markers at the proximal end of side and F2 group inoculation.Based on the group of F2 resistant strains which conform to 3:1,primers were designed to locate the invisible population.The results showed that the Fw3 was located between the marker ZJUd03067 and ZJUd03018 of chromosome A01,with the genetic distance between the two markers of 7.8 cM and 10.7 cM respectively.The interval contains 67 genes.This study provides a reference for cloning and molecular mechanism analysis of resistance genes of Fusarium wilt disease in sea-island cotton.
