照山白杜鹃愈伤组织诱导及植株再生技术 被引量:3

Callus Induction and Plant Regeneration Technology of Rhododendron micranthum

摘要 为了建立照山白杜鹃组培快繁技术体系,本研究以照山白杜鹃(Rhododendron micranthum)叶片及茎段为外植体,分析不同激素种类及浓度对愈伤组织诱导、不定芽生长及生根的影响。结果表明:TDZ与NAA组合有利于照山白杜鹃愈伤组织诱导,愈伤组织质量更优,愈伤组织诱导培养基的配方为WPM+1.0 mg/L TDZ+0.6 mg/L NAA,诱导率达96.30%。照山白杜鹃不定芽生长培养基的WPM+1.5 mg/L NAA+2.5 mg/L ZT,分化率达(89.5±1.2)%,增殖系数达4.9。适宜照山白杜鹃生根培养基为1/2WPM+0.8 mg/L IBA+0.25 mg/L NAA+0.5AC,生根率达(92.6±2.7)%。本研究建立了以照山白叶片和茎段为外植体的高效再生体系,对指导照山白优良种苗的离体快速繁殖具有重要意义,并为开展照山白的基因工程育种研究提供技术支撑。 In order to es tablish the tissue culture and rapid propagation technology system of Rhododendron micranthum,using the leaves and stems of Rhododendron micranthum as explants.The influence of different hormone types and concentration on callus induction,adventitious bud differentiation and root formation was studied.It was showed that TDZ and NAA were beneficial to the callus induction of Rhododendron micranthum and the callus tissue was easier to induce adventitious bud.The suitable medium for callus induction of Rhododendron micranthum was WPM+1.0 mg/L TDZ+0.6 mg/L NAA.The callus inductivity was 96.30%.The appropriate medium for adventitious buds differentiation was WPM+1.5 mg/L NAA+2.5 mg/L ZT,the differentiation rate reached(89.5±1.2)%,and the proliferation rate of adventitious buds reached 4.9.The suitable medium for rooting of Rhododendron micranthum was 1/2WPM+0.8 mg/L IBA+0.25 mg/L NAA+0.5 AC,and the rooting rate reached(92.6±2.7)%.In this study,a highly efficient regeneration system was established with the leaves and stems of Rhododendron micranthum as explants,which was of great significance for rapid propagation in vitro of Rhododendron micranthum seedlings,providing technical supports for genetic engineering of Rhododendron micranthum.
