2种包埋绿原酸产品体内外抗氧化能力的研究 被引量:3

Study on the anti-oxidation ability of two kinds of embedded chlorogenic acid products in vivo and in vitro

摘要 绿原酸(chlorogenic acid,CGA)具有良好的生物活性,具有抗氧化、抗炎、降低血脂、预防糖尿病和高血压等特性,但其还原性强,难以长期保存,生物利用度较低。为提高绿原酸利用度,延长其生物活性,前期制备2种绿原酸包埋物,分别为单独用多孔淀粉包埋绿原酸产品CGAP(绿原酸+多孔淀粉组)以及同时用多孔淀粉和卵磷脂共包埋绿原酸产品CGAPP(绿原酸+多孔淀粉+卵磷脂组),研究这2种绿原酸包埋物体内外抗氧化活性,并在体外测定包埋绿原酸随时间变化的稳定性。结果表明:这2种包埋工艺均能显著提高受到急性氧化损伤小鼠肝脏中超氧化物歧化酶和过氧化物酶活性,降低谷丙转氨酶活性和丙二醛含量。体外抗氧化试验表明,CGAPP的DPPH自由基清除率在0~80d时均保持在40%以上,超氧阴离子清除率在0~80d时均保持在15%以上;而CGAP在50d时体外抗氧化指标显著下降。CGAPP中绿原酸含量在80d内均保持在80%以上,CGAP中绿原酸含量为68.5%。2种包埋物绿原酸保存时间稳定性差异显著。这说明绿原酸+多孔淀粉+卵磷脂的包埋工艺在不影响绿原酸体内体外抗氧化能力情况下,能延长绿原酸的稳定性,为绿原酸的生产加工及保存提供了新思路。 Chlorogenic acid(CGA)has good biological activity,and is an important and biologically active dietary polyphenol.This polyphenol has many health-promoting properties,most of which are related to the treatment of metabolic syndrome,including antioxidant,anti-inflammatory,anti-lipid,anti-diabetic,and antihypertensive activities.Therefore,it has become an excellent candidate in the field of food additives and disease control.Because of its strong reducibility,it is difficult to store for a long time,and its bioavailability is relatively low.To improve the availability of chlorogenic acid and extend its biological activity,we prepared two kinds of chlorogenic acid embedding materials:chlorogenic acid+porous starch group and chlorogenic acid+porous starch+lecithin group.In this experiment,the antioxidant activity of these two chlorogenic acid-embedded objects was studied in vitro and in vitro,and the stability of the embedded chlorogenic acid over time was measured in vitro.The results showed that these two embedding processes can significantly increase the activity of superoxide dismutase and peroxidase in the liver of acute oxidative injury mice and reduce alanine aminotransferase activity and the content of malondialdehyde.In vitro,antioxidant experiments showed that the DPPH free radical scavenging rate of the CGAPP group remained above 40%,and the superoxide anion removal rate remained above 15%from 0-80 days.In the CGAP group the anti-oxidation indexes in vitro decreased significantly on the 50 th day.Finally,the residual rate of chlorogenic acid in the CGAPP group remained above 80%until 80 days,which was significantly higher than 68.5%of the CGAP group.These findings indicated that the embedding process of chlorogenic acid+porous starch+lecithin can maintain the scavenging efficiency of chlorogenic acid in vitro for DPPH free radicals and superoxide anion free radicals without affecting the ability of chlorogenic acid to resist oxidative stress in vivo.It also prolonged the stability of chlorogenic acid,which provides a new idea for the production,processing,and storage of chlorogenic acid.
