摘要
目的对木棉花水提物进行急性毒性试验,评价木棉花水提物的安全性,为后续的实验研究确定木棉花水提物用药剂量提供重要的实验依据。方法选择昆明小鼠32只,按完全随机法分为两组,即空白组和给药组各16只。给药组小鼠给予木棉花水提物最大给药量灌胃给药,约每12 h给药1次,1天内共给药剂量为120 g/kg。空白组小鼠给予与给药组等体积生理盐水灌胃,灌胃的次数同给药组。观察小鼠是否有死亡及其体重变化,并在给药14 d后通过颈椎脱臼将两组小鼠处死,观察重要脏器的变化。结果给药期间小鼠无死亡,仅体重有轻微的波动,但总体呈增长趋势。两组第1、5、10、14天体重分别比较,差异无统计学意义(P>0.05);肉眼观小鼠心、肝、脾、肺、肾、胸腺无明显出血、充血、水肿改变;两组肝脏、脾脏、胸腺、心脏、肺、左肾、右肾指数比较,差异无统计学意义(P>0.05)。结论木棉花水提物给药剂量为120 g/kg时,无明显毒性。
Objective To evaluate the safety of aqueous extract of Kapok by conducting an acute toxicity test,and to provide an important experimental basis for subsequent experimental studies to determine the dose of aqueous extract of Kapok.Methods Thirty-two Kunming mice were divided into two groups according to the complete randomization method,16 mice in the blank group and 16 mice in the drug-administered group.The mice in the drug-administered group were given the maximum dose of Kapok aqueous extract by gavage,approximately every 12 hours,for a total dose of 120 g/kg in a day,while the mice in the blank group were given the same volume of saline as the drug-administered group by gavage,and the number of gavage was the same as that of the drug-administered group.During the administration period,the mice were observed for death and changes in body weight,and after 14 days of administration,the mice in both groups were sacrificed by cervical dislocation to observe the changes in vital organs.Results There was no mortality in the mice during administration,only a slight fluctuation in the body weight of the mice and an overall increasing trend in the mice of the blank combination administration group.There was no significant difference in body weight between the two groups on day 1,5,10 and 14(P>0.05).The heart,liver,spleen,lung,kidney and thymus of mice had no obvious bleeding,hyperemia and edema changes in the naked eye.There was no significant difference in liver,spleen,thymus,heart,lung,left kidney and right kidney index between the two groups(P>0.05).Conclusion No significant toxicity was observed when the aqueous extract of Kapok was administered at a dose of 120 g/kg.
