摘要
目的:探讨Foxp3和SMARCE1在宫颈癌中表达水平及其相关临床意义。方法:选取60例宫颈癌组织作为研究对象,通过免疫组织化学检测癌组织中Foxp3和SMARCE1的表达水平,分析宫颈癌组织中Foxp3和SMARCE1的表达与患者临床病理资料的相关性;免疫组化检测SMARCE1在宫颈上皮内瘤变(CIN)组织中的表达;质粒构建及转染、Western blot、细胞免疫荧光、双荧光素酶、染色质免疫共沉淀等分子生物学实验检测Foxp3和SMARCE1表达的相关性。结果:Foxp3蛋白表达与宫颈癌更高的临床分期有关(P<0.05);SMARCE1蛋白与宫颈癌的淋巴结转移有关(P<0.05);Foxp3和SMARCE1蛋白的表达呈正相关(P<0.05);SMARCE1蛋白在宫颈高级别上皮内瘤变组织中的表达强于其在低级别上皮内瘤变(P<0.05);Foxp3可增强SMARCE1蛋白的表达,并可转录激活SMARCE1促进其表达。结论:宫颈癌中Foxp3和SMARCE1蛋白的表达成正相关,Foxp3可转录激活SMARCE1表达,参与宫颈癌的发生发展。
Objective:To investigate the expression levels of Foxp3 and SMARCE1 in cervical cancer and their clinical significance.Methods:The expression levels of Foxp3 and SMARCE1 in 60 cervical cancer tissues were detected by immunohistochemistry,and the correlation between the expression levels of Foxp3 and SMARCE1 in cervical cancer tissues and clinicopathological data of patients was analyzed.The expression of SMARCE1 in cervical intraepithelial neoplasia(CIN)was detected by immunohistochemistry.The correlation of Foxp3 and SMARCE1 expression was detected by molecular biology assay.Results:Foxp3 protein expression was associated with higher clinical stage of cervical cancer(P<0.05).SMARCE1 protein was associated with lymph node metastasis of cervical cancer(P<0.05).The expression of Foxp3 and SMARCE1 protein was positively correlated(P<0.05).The expression of SMARCE1 protein in high grade cervical intraepithelial neoplasia was stronger than that in low grade cervical intraepithelial neoplasia(P<0.05).Foxp3 could enhance the expression of SMARCE1 protein and transcriptionally activate SMARCE1 to promote its expression.Conclusion:Foxp3 and SMARCE1 protein expression are positively correlated in cervical cancer,Foxp3 can activate the expression of SMARCE1,and SMARCE1 is involved in the occurrence and development of cervical cancer.
