蓬莪术油微波提取工艺优化及抗氧化活性研究 被引量:6

摘要 目的:采用响应面法优化蓬莪术油微波辅助提取工艺,并研究蓬莪术油的抗氧化活性。方法:利用单因素试验考察微波提取温度、提取时间、料液比及提取功率对蓬莪术油得率影响后,采用响应面试验对微波辅助提取蓬莪术油的工艺进行优化,并通过对蓬莪术油还原性能力、清除DPPH·和·OH能力的测定初步评价其抗氧化活性。结果:蓬莪术油最佳提取条件:提取温度52℃、提取时间302s、液料比24.2∶1 mL/g,提取功率800W,在此条件下,蓬莪术油的得率为4.68%。当蓬莪术油浓度为200μg/mL时,对DPPH·清除率可达到70%,与VC对照品接近。结论:利用Box-Behnken响应面法优化得到的微波辅助提取蓬莪术油工艺稳定、可靠,可用于提取蓬莪术中挥发油,蓬莪术油具有较好的抗氧化活性。 Objective:Response surface methodology was used to optimize the microwave-assisted extraction process of the Curcuma kwangsiensis oil and the antioxidant activity of C.kwangsiensis oil was also studied.Methods:After investigating the effects of microwave extraction temperature,extraction time,material-to-liquid ratio and extraction power on the yield of C.kwangsiensis oil through single-factor experiments,the process of microwave-assisted extraction the C.kwangsiensis oil from C.kwangsiensis was optimized by response surface methodology,then the scavenging ability of DPPH· and ·OH was determed to preliminarily evaluated the antioxidant activity of C.kwangsiensis oil.Results:The optimum microwave assisted extraction process was as follows:52℃ of the extraction temperature,302s of the extraction time,24.2∶1mL/g of the liquid to material ratio,800 W of the extraction power.The yield was 4.68% under the optimum extraction conditions,when the concentration of C.kwangsiensis oil was 200μg/mL,the clearance rate of DPPH· could reach 70%,which was close to that of VC control.Conclusion:The microwave-assisted extraction process optimized by Box-Behnken response surface methodology is stable and reliable,and can be used to extract C.kwangsiensis oil from C.kwangsiensiss,C.kwangsiensis oil has good antioxidant activity.
