基于16S rDNA基因测序技术分析不同体长斑马鱼的肠道菌群结构差异 被引量:3

摘要 目的:分析探究同亲代不同体长斑马鱼肠道菌群组成和丰度差异。方法:将同一亲代的3月和6月龄子代斑马鱼,按性别及体长进行分组,采用16S rDNA序列分析技术检测斑马鱼肠道菌群构成,比较不同性别及体长的斑马鱼肠道菌群多样性和丰度差异。结果:Alpha多样性分析结果显示,3月龄斑马鱼同一性别体长与体短组间菌群丰度和多样性存在明显差异,生长至6月龄时,这一差异则无统计学意义。Beta多样性分析结果显示不同体长组肠道菌群具有明显的分群;6月龄优势菌门中,前6种优势菌门在雄性长组与短组组间差异无统计学意义;变形杆菌门和梭杆菌门在雌性长组与短组组间差异显著。线性判别分析结果显示,不同月龄体长长组与短组分别富集7个和3个菌门群落;不同月龄长组与短组分别富集13个和7个菌属群落。结论:斑马鱼体长与肠道菌群多样性密切相关,调节肠道菌群的结构及丰度可能是促进骨骼生长的有效途径。 Objective To analyze the composition and abundance of intestinal flora of zebrafish with different body lengths from the same parent.Methods Zebrafish offsprings aged 3 and 6 months from the same parental generation were divided into groups according to gender and body length.The composition of intestinal flora of zebrafish was detected by 16S rDNA sequence analysis,and the diversity and abundance of intestinal flora of zebrafish with different sex and body length were compared.Results The results of Alpha diversity analysis showed that there were significant differences in the abundance and diversity of bacterial flora between the 3-month-old zebrafish groups with the same sex and short body,but the difference was not statistically significant when the zebrafish grew to 6 months of age.The results of Beta diversity analysis showed that the intestinal flora of different body length groups had obvious grouping.Among the dominant bacteria of 6-month-old zebrafish,there was no significant difference between the male long group and the short group.There were significant differences in Proteobacteria and Fusobacteriota between the female long group and the short group.The results of linear discriminant analysis showed that 7 and 3 phyla communities were enriched in the long group and short group at different months of age,respectively.13 and 7 bacterial communities were enriched in the long and short groups of different months,respectively.Conclusion The body length of zebrafish is closely related to the diversity of intestinal flora.Regulating the structure and abundance of intestinal flora may be an effective way to promote skeletal development.
