大黄、赤芍注射液调控JAK/STAT信号通路促进急性肝衰竭大鼠肝再生的机制研究 被引量:5

摘要 观察大黄、赤芍注射液介导JAK/STAT信号通路在急性肝衰竭大鼠中的作用及对肝再生的影响。选用60%肝切除术后+20 mg/100g D-GalN+1μg/100 g LPS腹腔注射构建ALF大鼠模型,将造模成功的大鼠随机选取100只,按体质量随机均分为造模阳性对照组、促肝细胞生长素组、大黄、赤芍(低、中、高)剂量组。采用qPCR、Western Blot技术验证造模后24、48 h各组大鼠JAK2、JAK3、STAT3、STAT2表达。结果表明:大黄、赤芍注射液能够上调JAK2/JAK3、STAT2/STAT3蛋白表达,活化JAK/STAT信号通路,促进肝细胞再生,与对照组比较差异具有统计学意义P<0.05;其中低剂量组的各项检测结果优于(中、高)剂量组,差异具有统计学意义,P<0.05。大黄、赤芍注射液可改善急性肝衰竭大鼠肝功能,提高大鼠存活率,其机制可能与大黄、赤芍注射液降低炎症因子水平,活化JAK/STAT信号通路,诱导肝细胞再生有关。 To observe the effect of rhubarb and red peony injection on tyrosine kinase of JAK/STAT signaling and its effect on liver regeneration in rats with acute liver failure.60% intraperitoneal injection with + 20 mg/100 g D-GalN + 1 μg/100 g LPS. 100 rats were randomly divided into mold-positive control group, hepatocyte auxin group, low dose group, rhubarb and red peony according to body mass, High-dose group of red peony.Expression of JAK2, JAK3, STAT2, STAT3 at 24 h and 48 h after molding was verified by qPCR and Western Blot.rhubarb and red peony injections could upregulate JAK2/JAK3 and STAT2/STAT3 protein expression, activate JAK/STAT signaling pathway and promote hepatocyte regeneration with statistically significant P <0.05;the lower dose groups were better than the medium and higher doses, statistically significant and P <0.05. The injection of rhubarb and red peony can improve the liver function and survival rate of rats with acute liver failure. The mechanism may be related to the reduction of inflammatory factors and activation of JAK/STAT signaling and induction of hepatocyte regeneration.
