玉米肽对肝星状细胞LX2活化的作用

摘要 以玉米肽为研究对象,测定其氨基酸组成和相对分子质量分布,通过RT-PCR检测转化生长因子(transforming growth factor beta 1,TGF-β1)诱导人肝星状细胞LX2并经玉米肽干预后与肝纤维化相关基因的表达情况,并探讨玉米肽抗肝纤维化可能的作用机制。结果表明:玉米肽的氨基酸组成丰富,相对分子质量小于1 000的肽段含量高达95.68%;与正常组相比,模型组促进肝纤维化的TGF-β1、Ⅰ型胶原α1(COL1A1)、α-平滑肌肌动蛋白(α-SMA)、Smad2和Smad3基因的mRNA表达水平均极显著升高;与模型组相比,经不同质量浓度玉米肽干预后的LX2细胞,其TGF-β1、COL1A1、α-SMA、Smad2和Smad3基因的mRNA表达水平显著或极显著降低。说明玉米肽能够抑制人肝星状细胞的活化,降低LX2细胞中TGF-β1、COL1A1等促纤维化基因的表达,其分子机制可能与抑制TGF-β1/Smad信号通路活性有关。 Using corn peptide as the research object,the amino acid composition and relative molecular mass distribution were determined.Reverse transcription-polymerase chain reaction(RT-PCR) was used to detect the expression of genes related to liver fibrosis induced by transforming growth factor beta 1(TGF-β1) in human hepatic stellate cells and intervened by corn peptide,and to explore the possible mechanism of anti-hepatic fibrosis.The results showed that the corn peptides were rich in amino acid composition,and the content of peptides with relative molecular mass less than 1 000 was as high as 95.68 %.Compared with the normal group,the expression levels of TGF-β1,COL1 A1,α-smooth muscle actin(α-SMA),Smad2 and Smad3 genes in the model group were significantly increased.Compared with model group,mRNA expression levels of TGF-β1,COL1 A1,α-SMA,Smad2 and Smad3 genes were significantly or extremely significantly decreased in LX2 cells treated with different mass concentrations of corn peptides.It was concluded that corn peptide could inhibit the activation of human hepatic stellate cells and decrease the expression levels of TGF-β1,COL1 A1 and other profibrotic genes in LX2 cells and the molecular mechanism might be related to the inhibition of TGF-β1/Smad signaling pathway activity.
