改良型富血小板纤维蛋白用于口腔软组织缺损修复对瘢痕的影响 被引量:3

摘要 目的探讨改良型富血小板纤维蛋白(APRF)修复兔硬腭软组织缺损中对瘢痕组织的作用机制。方法将30只日本大耳白兔随机分为两组,于APRF组的兔硬腭软组织缺损处植入自体APRF膜,空白对照组缺损处自然愈合。建模后3、7、14、21、28 d观察术区瘢痕形成情况,通过Masson染色及酶联免疫吸附法(ELISA)行羟脯氨酸及2种生长因子[血小板衍生生长因子(PDGF)、转化生长因子β(TGF-β)]定量分析,比较二者之间的差异。结果(1)空白对照组缺损在实验结束时自行愈合,但瘢痕明显;(2)建模后3、7、14、21、28 d标本的大体观察、Masson染色、ELISA均显示APRF组在胶原合成与降解方面均明显优于空白对照组,差异均有统计学意义(P<0.05)。结论改良型富血小板纤维蛋白在促进软组织愈合中的胶原合成与降解方面能力较优,能够较好地减少瘢痕组织的形成。 Objective To explore the mechanism of advance platelet-rich fibrin(APRF)on scar tissue in repairing soft tissue defects of hard palate in the rabbits.Methods A total of 30 Japanese rabbits with large ears were randomly divided into two groups.The soft tissue defects of the hard palate were implanted with autologous APRF membrane in the experimental group(APRF group),and the defects of the blank control group were healed naturally.At 3,7,14,21 and 28 days after the modeling,the scar formation in the operative area was observed.Masson staining and enzyme linked immunosorbent assay(ELISA)were used for the quantitative analysis of hydroxyproline and other two growth factors(platelet-derived growth factor,PDGF;transforming growth factor β,TGF-β).The differences were compared between the two groups.Results(1)Defects in the blank control group healed spontaneously at the end of the experiment,but the scar was obviously;(2)Gross observation of 3,7,14,21 and 28 days after the modeling,the Masson staining and ELISA showed that the APRF group was significantly better than the blank group in terms of the collagen synthesis and degradation,and the differences were statistically significant(P<0.05).Conclusion APRF had better ability in promoting collagen synthesis and degradation in the soft tissue healing.It could well reduce the formation of scar tissue.
