基于单细胞测序数据系统分析SARS-CoV-2受体ACE2在胰腺中的表达

摘要 目的通过单细胞测序数据系统分析严重急性呼吸系统综合征冠状病毒2(SARS-CoV-2)受体ACE2在胰腺中的表达,为新型冠状病毒感染性疾病(COVID-19)患者提供了生物信息学证据。方法利用单细胞表达谱数据库中的人类胰腺组织单细胞测序数据用于ACE2表达阳性细胞的聚类分析。Scanpy软件用于分析scRNA序列的下游数据。Seurat R软件包用于原始基因表达矩阵的进一步数据分析。然后进行功能富集分析,并使用可视化的R软件对ACE2阳性标记基因进行了基因概念网络分析。结果ACE2主要分布于胰腺腺泡细胞和导管内皮细胞,ACE2阳性细胞在与膜靶向蛋白、胞浆核糖体和内质网蛋白定位建立相关的病毒翻译相关基因集中高度富集。ACE2阳性腺泡细胞中的标记基因与各种类型胰腺炎的发生最为相关,而ACE2阳性胰腺导管内皮细胞中的标记基因与癌前病变密切相关。结论研究结果表明,人胰腺外分泌腺是SARS-CoV-2感染的潜在靶器官,可能对这种快速且高度传播的疾病的病理生理学理解产生重大影响。 Objective To study the expression of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)receptor angiotensin-converting enzyme 2(ACE2)in pancreas by single-cell sequencing data systems,and to provide bioinformatics evidence for patients with Corona Virus Disease 2019(COVID-19).Methods The single cell sequencing data of human pancreas tissue in the single cell expression profile database was used for cluster analysis of ACE2-expressing positive cells.Scanpy software was used to analyze the downstream data of the scRNA-seq.The Seurat R software package was used for further data analysis on the raw gene expression matrix.Then functional enrichment analysis was performed,and the gene concept network analysis of ACE2 positive marker genes was performed by using visual R software.Results ACE2 was mainly distributed in pancreatic acinar cells and ductal endothelial cells,and the ACE2 positive cells were highly enriched in viral translation-related genes related to the establishment of membrane-targeted protein,cytoplasmic ribosome and endoplasmic reticulum protein localization.Marker genes in ACE2-positive acinar cells were most related to the occurrence of various types of pancreatitis,while marker genes in ACE2-positive pancreatic duct endothelial cells were closely related to precancerous conditions.Conclusion The results suggest that the human pancreatic exocrine gland is a potential target for SARS-CoV-2 infection,which may have a significant impact on the understanding of the pathophysiology of this rapid and highly transmitted disease.
