摘要
目的 评价芩柏软膏、芩柏凝胶、芩柏乳膏体外经皮渗透特性及其抗银屑病作用。方法 采用改良的Franz扩散池法进行体外经皮渗透实验,HPLC法测定1、2、4、6、8、10、12 h经正常、模型皮肤接收液中黄芩苷、盐酸小檗碱含量。建立咪喹莫特诱导的银屑病样小鼠模型,考察3种芩柏制剂对小鼠皮损的影响。结果 芩柏凝胶中黄芩苷、盐酸小檗碱累积渗透量和皮肤储药量最高,芩柏软膏中最低,正常皮肤中上述指标高于模型皮肤中。3种芩柏制剂体外经皮渗透均符合零级动力学方程。与模型组比较,3种芩柏制剂组小鼠皮损改善,表皮厚度减少(P<0.01)。结论 3种芩柏制剂具有抗银屑病作用,其中芩柏凝胶中黄芩苷、盐酸小檗碱渗透速率、累积渗透量、皮肤储药量最高。
AIM To evaluate the in vitro percutaneous permeation characteristics and anti-psoriatic effects of Qinbai Ointment,Qinbai Gel and Qinbai Cream.METHODS In vitro percutaneous permeation test was performed by modified Franz diffusion cell method.HPLC was adopted in the content determination of baicalin and berberine hydrochloride in receiving fluid through normal skins and model skins at 1,2,4,6,8,10,12 h.The imiquimod-induced mouse psoriasis-like model was established,after which the effects of three Qinbai preparations on mouse skin lesions were investigated.RESULTS The accumulative permeation amount and storage amount of baicalin and berberine hydrochloride were the highest in Qinbai Gel and the lowest in Qinbai Ointment,and the aforementioned indices in the normal skins were higher than those in the model skin.The in vitro percutaneous permeation of three Qinbai preparations accorded with zero-order kinetic equation.Compared with the model group,three Qinbai preparation groups demonstrated improved mouse skin lesions and reduced epidermal thickness(P<0.01).CONCLUSION Three Qinbai preparations exhibit anti-psoriasis effects,among which baicalin and berberine hydrochloride in Qinbai Gel display the highest permeation rate,accumulative permeation amount and skins drug storage amount.
