姜黄素对牙周炎大鼠模型骨丢失的影响及相关机制研究

摘要 目的探讨与研究姜黄素对牙周炎大鼠模型骨丢失的影响及相关机制。方法选取50只清洁级雄性Wistar大鼠,2~3月龄,体重180~220 g。建模过程中死亡2只,建模成功48只。将牙周炎大鼠模型随机数字表法分为3组-模型组、姜黄素1组、姜黄素2组,每组各16只。姜黄素1组、姜黄素2组按50、100 mg/kg给予灌服姜黄素,模型组给予灌服等剂量的0.9%氯化钠溶液,1次/d。治疗第15天与第30天,采用酶联免疫吸附试验法检测血清肿瘤坏死因子(TNF)-ɑ和白细胞介素(IL)-1β表达水平;测定与记录骨小梁间隙、骨小梁数目、骨小梁厚度;记录所有大鼠的釉牙骨质界到牙槽嵴顶的距离,计算牙槽骨吸收体积;采用蛋白质印迹法检测骨保护素(OPG)与核因子-κB受体活化因子配体(RANKL)蛋白相对表达水平。结果治疗第15天与第30天,姜黄素1组、姜黄素2组的血清TNF-ɑ、IL-1β表达水平低于模型组,差异均有统计学意义(P<0.05)。治疗第15天与第30天,姜黄素1组、姜黄素2组的骨小梁数目、骨小梁厚度高于模型组,骨小梁间隙低于模型组,差异均有统计学意义(P<0.05)。治疗第15天与第30天,姜黄素1组、姜黄素2组的釉牙骨质界到牙槽嵴顶距离与牙槽骨吸收体积低于模型组,差异均有统计学意义(P<0.05)。治疗第15天与第30天,姜黄素1组、姜黄素2组的OPG与RANKL蛋白相对表达水平低于模型组,差异均有统计学意义(P<0.05)。且姜黄素2组的作用优于姜黄素1组。结论姜黄素在牙周炎大鼠模型的应用能下调OPG/RANKL信号通路的表达,抑制炎症因子的释放,从而能缓解骨丢失情况,发挥骨保护作用。 Objective To explore and study the effect of curcumin on bone loss in a rat model of periodontitis and related mechanisms.Methods Fifty clean-grade male Wistar rats,2-3 months old,weighing 180-220 g were selected.During the modeling process,2 died,and 48 were successfully modeled.Periodontitis rat models were randomly divided into three groups-model group,curcumin 1 group,curcumin 2 group,each group 16 rats.The curcumin 1 group,curcumin 2 groups were given curcumin at 50,100 mg/kg,and the model group were given the same dose of normal saline once daily.On the 15th and 30th days of treatment,the expression levels of serum tumor necrosis factor(TNF)-ɑand interleukin(IL)-1βwere detected by enzyme-linked immunosorbent assay;trabecular bone space,trabecular bone number,bone trabecular thickness was measured and recorded;the distance from the cementoenamel junction to the alveolar crest was recorded in all rats,and the alveolar bone resorption volume was calculated;the relative expression level of osteoprotegerin(OPG)and nuclear factor-κB receptor activator ligand(RANKL)protein was detected by Western blotting.Results On the 15th and 30th day of treatment,the expression levels of serum TNF-ɑand IL-1βin curcumin group 1 and curcumin group 2 were lower than those in model group,and the differences were statistically significant(P<0.05).On the 15th and 30th day of treatment,the number of trabecular bone and thickness of trabecular bone in curcumin group 1 and curcumin group 2 were higher than those in the model group,and the trabecular bone gap was lower than that in the model group,and the differences were statistically significant(P<0.05).On the 15th and 30th day of treatment,the distance from cementumenamel junction to alveolar crest and alveolar bone resorption volume in curcumin group 1 and curcumin group 2 were lower than those in model group,and the differences were statistically significant(P<0.05).On the 15th and 30th days of treatment,the relative expression levels of OPG and RANKL proteins in the curcumin group 1 and curcumin group 2 were lower than those in the model group,and the differences were statistically significant(P<0.05).And the effect of curcumin group 2 was better than that of curcumin group 1.Conclusion The application of curcumin in a rat model of periodontitis can down-regulate the expression of OPG/RANKL signaling pathway and inhibit the release of inflammatory factors,thereby alleviating bone loss and exerting bone protection.
