摘要
以表皮有刺毛和无刺毛的2种佛手瓜为研究材料,对其SeTTG1转录因子进行克隆和分析。结果显示:2种佛手瓜中SeTTG1转录因子的ORF框长度均为1026 bp,可编码341个氨基酸。无刺毛和有刺毛佛手瓜的SeTTG1核苷酸序列5个位点存在差异,分别在421、466、594、683和780位点上。推导的氨基酸存在3个位点差异,分别在141、156和228位点上。2种佛手瓜SeTTG1转录因子均含4个WD40基序。进化上,佛手瓜SeTTG1与葫芦科植物TTG1蛋白同源性较高。有刺佛手瓜茎和瓜中SeTTG1表达分别显著(P<0.05,1.36倍)和极显著(P<0.01,35.64倍)高于无刺瓜,而根和叶中的表达水平在2种类型中无显著差异。
In this study,transcription factors SeTTG1 were isolated and studied from two genotypes of S.edule with or without peel hair.The results showed that the ORF of SeTTG1 was 1026 bp,which could encode 341 amino acid residues in two genotypes of S.edule.There were five differences in SeTTG1 nucleotide sequences of hairless and hairs S.edule,which was 421,466,594,683 and 780 site,respectively.The deduced amino acids were different in three sites:141,156 and 228,respectively.The sequence of SeTTG1 contained four WD40 motifs.Evolutionarily,SeTTG1 had high homology with TTG1protein of other cucurbitaceae.The expression level of SeTTG1 in stem and fruits of peel hair of S.edule was significantly and extremely significant higher than that of peel hairless,respectively,while there was no significant difference in roots and leaves in two genotypes S.edule.
