一株猪伪狂犬病病毒的分离鉴定及其对不同动物的致病性 被引量:1

摘要 为了解变异后伪狂犬病病毒(Pseudorabies virus,PRV)的致病性,采用细胞培养技术从病料中分离PRV,通过PCR方法对PRV相关基因进行扩增,对毒株进行特异性试验和病毒滴度的测定,并通过动物试验对PRV的致病性进行了研究。结果表明:通过PCR可扩增出大小与预期相符的PRV gG基因片段、gE基因片段以及TK基因片段;病毒毒价可达10^(8.0) TCID_(50)∕mL;接种10^(5.0)TCID_(50)∕mL病毒后,家兔于52 h左右全部死亡,小鼠于72 h左右全部死亡,10^(4.0) TCID_(50)∕mL病毒可致28日龄仔猪死亡或发病。本研究可为预防PRV疫病的流行提供防控依据。 In order to understand the pathogenicity of pseudorabies virus(PRV)after mutation,a PRV was isolated from the disease material by cell culture technology.The PRV related genes were amplified by PCR,the specific test of the virus and the determination of virus titer were carried out,and the pathogenicity of PRV was studied by animal test.The results showed that gG gene fragment,gE gene fragment and TK gene fragment of PRV with the expected size could be amplified by PCR.The virus titer could reach 108.0 TCID_(50)∕mL.After inoculation with 10^(5.0)TCID_(50)∕mL virus,all rabbits died in about 52 hours,all mice died in about 72 hours,and 10^(4.0) TCID_(50)∕mL virus could cause the death or disease of 28 day old piglets.This study could provide basis for prevention and control of PRV epidemic.
