摘要
目的利用高通量基因芯片技术检测老年心力衰竭患者血浆中环状RNA(cirRNAs)的表达谱,进行生物信息学分析,探究血浆环状RNA在老年心力衰竭发生发展中的作用。方法选取安徽省立医院确诊为老年心力衰竭患者4例(研究组)和无心血管疾病的老年人4例(对照组)作为研究对象,运用基因芯片技术检测2组对象血浆中环状RNA表达谱,筛选出2组间差异表达的环状RNA,对差异表达的环状RNA进行分析以及对其可能结合的微小RNA(miRNA)进行预测。结果与对照组比较,研究组血浆中差异表达的环状RNA共190种(变化倍数>1.5,P<0.05),其中表达上调的有116种,下调的有74种。其中可能与差异性表达的环状RNA结合的miR-764、miR-410-3p、miR-483-3p、miR-148b-3p、miR-204-5、miR-17-3p、miR-134-5p可能参与心力衰竭的病理过程。结论研究组与对照组血浆间存在差异表达的环状RNA,这些环状RNA可能参与老年心力衰竭的发生发展过程。
Objective To detect the expression profile of plasma circular RNA in elderly patients with heart failure,and explore the role of plasma circular RNA in the occurrence and development of senile heart failure.Methods Four elderly patients with heart failure(experimental group)and four elderly patients without cardiovascular disease(control group)were selected from Anhui Provincial Hospital.The expression profile of circular RNA in plasma of the two groups was detected by gene chip technology to screen the differentially expressed circular RNA between the two groups.The differentially expressed circular RNA were analyzed and their possible binding microRNAs(miRNA)were predicted.Results Compared with the control group,there were 190 differentially expressed circular RNA in the plasma of the experimental group(multiple of change>1.5,P<0.05),of which 116 were up-regulated and 74 were down-regulated.Among them,mir-764,mir-410-3p,mir-483-3p,mir-148b-3p,mir-204-5,mir-17-3p,and mir-134-5p that may bind to differentially expressed circular RNA were likely to be involved in the pathological process of heart failure.Conclusions There are differentially expressed plasma circular RNA between the experimental group and the control group,which may be involved in the occurrence and development of senile heart failure.
