摘要
目的:研究BMAP-28对乳腺癌Bcap37细胞增殖及凋亡的影响。方法:体外培养Bcap37细胞,设置对照组(BMAP-28浓度为0μg/mL)和实验组(BMAP-28浓度分别为20、40、60、80μg/mL)。CCK-8实验检测Bcap37细胞的增殖情况;DAPI染色观察细胞凋亡形态学变化;流式细胞术检测细胞凋亡率。结果:CCK-8实验测得BMAP-28对Bcap37细胞的增殖具有抑制作用,BMAP-28的IC_(50)=49.19μg/mL。DAPI染色可见随着BMAP-28浓度的增加,细胞凋亡数量增加,凋亡形态明显。流式细胞术显示细胞凋亡率逐渐增高,趋势和DAPI染色结果一致(P<0.01)。结论:BMAP-28能显著抑制乳腺癌Bcap37细胞的增殖,促进癌细胞的凋亡,有望成为临床乳腺癌治疗的新型抗癌药物。
Objective:To study the effects of BMAP-28 on the proliferation and apoptosis of breast cancer Bcap37 cells.Method:Bcap37 cells were cultured in vitro,the control group(BMAP-28 concentration was 0μg/mL)and the experimental group(BMAP-28 concentrations were 20,40,60,80μg/mL)were set up.CCK-8 assay was used to detect the cell proliferation of breast cancer Bcap37 cells,DAPI staining was used to observe the morphological changes of cell apoptosis,flow cytometry was used to detect the apoptosis rate of cells.Result:CCK-8assay showed that BMAP-28 can inhibit the proliferation of breast cancer Bcap37 cells,IC_(50)=49.19μg/mL.DAPI staining showed that with the increase of BMAP-28 concentration,the number of cell apoptosis increased,and the apoptotic morphology was obvious.Flow cytometry showed that the rate of cell apoptosis increased and the trend was consistent with the result of DAPI staining(P<0.01).Conclusion:BMAP-28 can significantly inhibit the proliferation of breast cancer Bcap37 cells and promote the apoptosis of cancer cells,which is hopefully to be a new type of anticancer agents for clinical breast cancer treatment.
