陕西汉中不同产区泥附子中酯型生物碱类成分的测定 被引量:1

Determination of ester alkaloids in aconite from different producing areas in Hanzhong,Shaanxi Province

摘要 目的以产自陕西汉中4个县(区)的15批泥附子为研究对象,采用超高效液相色谱法(ultra performance liquid chromatography,UPLC)同时测定苯甲酰新乌头原碱、苯甲酰次乌头原碱、苯甲酰乌头原碱、新乌头碱、次乌头碱和乌头碱6种酯型生物碱的含量。方法采用ACQUITY UPLC BEH色谱柱C_(18)(50 mm×2.1 mm,1.7μm);流动相:乙腈为流动相A,0.04 mol·L^(-1)乙酸铵溶液(氨水调pH=10)为流动相B;柱温:25℃;检测波长:235 nm。结果所测泥附子中的6种酯型生物碱分离度良好,15批样品中6种酯型生物碱的含量有明显差异。结论该方法可同时测定附子中6种酯型生物碱的含量,在泥附子与炮制品的质量评价中具有一定的参考价值。 Objective By taking 15 batches of aconite in 4 counties(districts)in Hanzhong,Shaanxi Province as the research targets,to simultaneously determine the contents of 6 ester-type alkaloids,benzoylmesaconine,benzoylaconine,benzoylhypacoitine,aconitine,hypaconitine,and mesaconitine.Methods ACQUITY UPLC BEH C_(18) column(50 mm×2.1 mm,1.7μm)was used with the mobile phase,A acetonitrile,B 0.04 mol·L^(-1) ammonium acetate solution(with ammonia water adjusting pH=10).The column temperature was 25℃ and the detection wavelength was 235 nm.Results The separation of the 6 ester-type alkaloids in the tested Aconite vulgaris was good,and the content of the 6 ester-type alkaloids in 15 samples had obvious differences.Conclusion The method has good precision and reproducibility.It can simultaneously determine the content of the 6 ester-type alkaloids in aconite,which can provide scientific reference for the quality evaluation of aconite and processed products.
